Tris(1,3-dichloro-2-propyl) phosphate reduces longevity through a specific microRNA-mediated DAF-16/FoxO in an unconventional insulin/ insulin-like growth factor-1 signaling pathway

Chen Wang et al.

Summary: This study investigated the harmful effects of TDCPP on longevity regulation signal pathways and biological processes in C. elegans, focusing on microRNAs (miRNAs) using small RNA transcriptome sequencing. Results suggested that TDCPP reduced nematode lifespan and mobility by up-regulating miR-48 and miR-84 to silence key regulators of longevity, ultimately providing a comprehensive analysis of TDCPP-reduced longevity from a miRNA perspective.

MicroRNA-7 overexpression positively regulates the CD8+ SP cell development via targeting PIK3R1

Lin Hu et al.

Summary: miR-7, a distinct member of miRNA family, has been found to play a crucial role in promoting the development of CD8(+) SP cells. Overexpression of miR-7 led to increased volume, weight and total cell numbers of thymus in mice. The enhancement of CD8(+) SP cell activation and proliferation was also observed in miR-7 overexpression mice, suggesting its intrinsic role in promoting CD8(+) T cell development. Furthermore, the regulation of PIK3R1 expression and the balance of AKT and ERK pathways were identified as potential mechanisms by which miR-7 promotes CD8(+) SP cell development.

NAD+ metabolism and its roles in cellular processes during ageing

Anthony J. Covarrubias et al.

Summary: NAD(+) is a crucial coenzyme in redox reactions and plays a pivotal role in energy metabolism. Its levels decline with age, and raising NAD(+) levels is being explored as a potential strategy to extend human lifespan and healthspan. NAD(+) influences key cellular processes and functions, including metabolic pathways, DNA repair, and cellular senescence, with implications for healthy aging and age-related diseases.

Co-expression analysis identifies networks of miRNAs implicated in biological ageing and modulated by short-term interval training

Prasun Kumar Dev et al.

Summary: Exercise training is associated with aerobic fitness and leukocyte telomere length in a positive manner, while vigorous exercise such as sprint interval training (SIT) can regulate miRNA networks, down-regulate immune response-related miRNAs clusters, and potentially attenuate biological ageing through controlling specific miRNA families like let-7 and miR-320.

Age-related increase of let-7 family microRNA in rat retina and vitreous

Priscilla Sayami Akamine et al.

Summary: The study found expression of the let-7 miRNA family in the vitreous and retina of rats, with most let-7 members significantly upregulated in the vitreous during development. Only let-7b and let-7c showed similar expression patterns in the retina as well. The increase of let-7b and let-7c in aging vitreous, along with their targeting of Has2 mRNA, may play a role in modulating hyaluronan content during the aging process.

Summary: Parkinson's Disease is a common neurodegenerative disease among older adults, characterized by dopaminergic neuron loss and abnormal accumulation of alpha-synuclein. The development of PD involves various factors such as genetic mutations, post-translational modifications, and mitochondrial dysfunction. The overproduction of reactive oxygen species activates alpha-synuclein aggregation, leading to neurodegeneration.

Targeting immune dysfunction in aging

Simone Borgoni et al.

Summary: Human aging is a multifactorial phenomenon that affects various organ systems and cellular processes, with the immune system being one of the most dysregulated. The immune changes that occur during aging, such as immunosenescence and inflammaging, result in increased susceptibility to infections, reduced healing abilities, and the emergence of age-associated diseases. Therapeutic approaches targeting specific immune subpopulations affected by aging have the potential to rejuvenate the immune system and improve healthspan.
