Exploiting senescence for the treatment of cancer

Liqin Wang et al.

Summary: This review discusses how senescence can be induced in cancer cells and how the distinctive features of senescent cancer cells can be exploited for selective eradication. It also explores activation of the host immune system as an attractive way to clear senescent cancer cells, and the potential benefits of a sequential treatment approach with pro-senescence therapy followed by senolytic therapy.

Stathmin dynamics modulate the activity of eribulin in breast cancer cells

Mikihiro Yoshie et al.

Summary: The study investigated the role of stathmin in the antiproliferative activity of eribulin in breast cancer cells. The results suggest that stathmin dynamics are closely associated with the antiproliferative effects of eribulin and stathmin is a possible biomarker for predicting the therapeutic effects of eribulin in breast cancer patients.

The PGRMC1 Antagonist AG-205 Inhibits Synthesis of Galactosylceramide and Sulfatide

Lihua Wang-Eckhardt et al.

Summary: The study found that AG-205 not only inhibits PGRMC1, but also affects other enzymes, indicating caution in its use. Experimental results showed that AG-205 primarily inhibits UDP-galactose: ceramide galactosyltransferase, rather than cerebroside sulfotransferase.

Deciphering the Role of PGRMC1 During Human Decidualization Using an In Vitro Approach

Stefania Salsano et al.

Summary: This study found that PGRMC1 inhibition during decidualization in endometrial stromal cells does not interfere with cytoskeletal rearrangements and prolactin secretion, but does affect the expression of genes related to metabolism, transport, and hormonal biosynthesis. Additionally, activation of membrane progesterone receptor signaling alone is insufficient to induce proper decidualization.

AG-205 Upregulates Enzymes Involved in Cholesterol Biosynthesis and Steroidogenesis in Human Endometrial Cells Independently of PGRMC1 and Related MAPR Proteins

Charlotte Thieffry et al.

Summary: The study showed that AG-205 affects gene expression in endometrial cells, but not through PGRMC1 or other MAPRs. AG-205 lacks specificity and its mechanisms of action are unclear, raising the risk for endometrial pathologies related to hormone imbalance.

Senolytic treatment modulates decidualization in human endometrial stromal cells

Kazuya Kusama et al.

Summary: Decidualization, a crucial step for embryo implantation and placentation, can be enhanced by senolytic agents such as Quercetin and Dasatinib. Treatment with a combination of these agents synergistically increases the expression of decidualization markers and senescence markers in decidualizing ESCs, offering a potential therapeutic strategy for infertility related to endometrial receptivity and embryo implantation dysfunctions.

Modelling the impact of decidual senescence on embryo implantation in human endometrial assembloids

Thomas M. Rawlings et al.

Summary: The study suggests that decidual senescence controls endometrial fate decisions at implantation and the presence of senescent decidual cells creates a dynamic implantation environment, enabling embryo expansion and attachment but leading to gradual disintegration of assembloids. Pharmacological inhibition of stress responses in pre-decidual cells accelerated decidualization and eliminated the emergence of senescent decidual cells, resulting in a strong, static decidual matrix. The research highlights how endometrial assembloids may facilitate the discovery of new treatments to prevent reproductive failure.
