TET2 mutations as a part of DNA dioxygenase deficiency in myelodysplastic syndromes

Summary: Decreased DNA dioxygenase activity from the TET2 gene family is important in MDS. The downregulation of 5-hydroxymethylcytosine (5-hmC) suggests a role for DNA demethylation in MDS beyond TET2 mutations. TET3 is upregulated in MDS as a compensatory mechanism for TET2 dysfunction, and elevated TET3 levels influence the clinical phenotype of TET2 deficiency.
