FBXO38 Ubiquitin Ligase Controls Sertoli Cell Maturation

Nikol Dibus et al.

Summary: The ubiquitin ligase SCFFBXO38 regulates centromeric chromatin by promoting the degradation of the ZXDB protein. Loss of FBXO38 leads to growth retardation, pathological changes in the testes, decreased sperm production, and reduced fertility in mice. FBXO38 is specifically expressed in Sertoli cells in the testes, and its absence results in Sertoli cell maturation defect and impaired spermatogonial differentiation.

Centromeres are dismantled by foundational meiotic proteins Spo11 and Rec8

Haitong Hou et al.

Summary: The study reveals that two key meiosis-defining proteins Spo11 and Rec8 can dismantle centromeres in the nucleus, posing a threat to genome stability. This dismantlement is typically observed in mutant cells lacking the telomere bouquet, indicating a potential danger in cancers expressing meiotic proteins.

CENP-A overexpression promotes distinct fates in human cells, depending on p53 status

Daniel Jeffery et al.

Summary: The study establishes an inducible and reversible CENP-A overexpression system, combined with a switch in p53 status in human cell lines, revealing p53 as a key determinant of how CENP-A impacts cell state, cell identity, and therapeutic response. The findings show that CENP-A overexpression promotes senescence and radiosensitivity when p53 is functional, while promoting epithelial-mesenchymal transition when p53 is inactivated.

CRL4-DCAF12 Ubiquitin Ligase Controls MOV10 RNA Helicase during Spermatogenesis and T Cell Activation

Tomas Lidak et al.

Summary: This study identified MOV10 as a novel substrate of CRL4-DCAF12 and demonstrated the biological relevance of the DCAF12-MOV10 pathway in spermatogenesis and T cell activation. Knockout of Dcaf12 in mice led to decreased sperm production and altered T cell populations, highlighting the importance of DCAF12 in reproductive and immune functions. Additionally, in vitro experiments showed that Dcaf12-deficient T cells displayed abnormal stability of MOV10 and increased levels of activated caspases.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

Aneuploidy as a promoter and suppressor of malignant growth

Summary: Aneuploidy, although frequent in human tumors, is not always a driver of cancer development and can have tumor-suppressive effects. The specific consequences of aneuploidy are context-dependent and influenced by genetic and environmental factors.
