Targeting the IspD Enzyme in the MEP Pathway: Identification of a Novel Fragment Class

Eleonora Diamanti et al.

Summary: The MEP pathway enzymes are crucial for synthesizing isoprenoids and are potential drug targets. A high-throughput screening approach led to the discovery of a novel fragment hit against PfIspD, and a systematic SAR investigation identified a new compound for structure-guided optimization. A putative binding mode for the inhibitors was proposed using a homology model of PfIspD.

Determination of up to twenty carboxylic acid containing compounds in clinically relevant matrices by o-benzylhydroxylamine derivatization and liquid chromatography-tandem mass spectrometry.

Alex Gomez-Gomez et al.

Summary: A LC-MS/MS method for the quantification of 20 endogenous R-COOH in different pathways was developed and optimized. The method was applied in various matrices and proved suitable for studying R-COOH status in health and disease through targeted metabolomics. Adequate accuracy and precision were achieved, making this methodology a valuable tool for biochemical research.

Triple Artemisinin-Based Combination Therapies for Malaria - A New Paradigm?

Rob W. van der Pluijm et al.

Summary: Recent progress in the fight against malaria is at risk due to the emergence and spread of artemisinin and partner drug resistance, particularly in the Greater Mekong Subregion. New drugs are not expected in the near future, making it crucial to utilize triple artemisinin-based combination therapies to combat falciparum malaria effectively. Deployment of such therapies beyond the GMS could potentially help prevent global resistance to currently available drugs.

Zwitterionic self-assembled nanoparticles as carriers for Plasmodium targeting in malaria oral treatment

Arnau Biosca et al.

Summary: Novel nanoparticles were designed and synthesized to enhance the bioavailability of antimalarials, showing specific targeting to Plasmodium falciparum-infected red blood cells with maintained in vitro efficacy upon encapsulation of the lipophilic antimalarial drug curcumin. In vivo assays demonstrated increased curcumin concentration in mouse blood after oral administration of the nanoparticles, leading to similar antimalarial activities compared to free drug in Plasmodium yoelii yoelii-infected mice. Microscopic analysis revealed the presence of the polymer inside parasitized erythrocytes one hour after oral administration to infected animals.

Targeted metabolomics in formalin-fixed paraffin-embedded tissue specimens: Liquid chromatography-tandem mass spectrometry determination of acidic metabolites in cancer research

Alex Gomez-Gomez et al.

Summary: Formalin-fixed paraffin-embedded (FFPE) tissues are crucial in cancer research, but research on the detection of small molecules in FFPE samples is limited. This study aimed to explore the potential of targeted metabolomics in FFPE specimens and developed a LC-MS/MS method for the quantification of acidic metabolites. The method showed accurate and precise quantification of 40 metabolites in FFPE samples, with implications for cancer research.

Protein Prenylation and Hsp40 in Thermotolerance of Plasmodium falciparum Malaria Parasites

Emily S. Mathews et al.

Summary: During the malaria parasite's life cycle, farnesylation plays a crucial role in its survival through temperature changes. Loss of farnesylation of HSP40 affects its membrane attachment and interaction with essential proteins, impacting the parasite's ability to survive.

The apicoplast link to fever-survival and artemisinin-resistance in the malaria parasite

Min Zhang et al.

Summary: A large-scale forward genetic screen in malaria-causing Plasmodium falciparum identified over 200 mutants with differential responses to increased temperature, with implications for artemisinin sensitivity and oxidative stress. Critical processes associated with parasite tolerance to fever and artemisinin resistance include protein-folding, heat shock, and isoprenoid biosynthesis pathways derived from the parasite's algal endosymbiont ancestor genome. Parasites appear to exploit their innate febrile response mechanisms for artemisinin resistance, involving genes linked to the evolutionary origins of Plasmodium parasites.
