Summary: CAT3 is a novel oral anti-glioma pro-drug with potent anti-tumor effects. However, its active metabolite PF403 causes significant gastrointestinal side effects. By using a self-microemulsifying drug delivery system to modify CAT3 metabolism, the bioavailability of CAT3 can be increased while reducing the generation of PF403, leading to improved anti-glioma effects and reduced gastrointestinal side effects simultaneously.

Brain penetrating peptides and peptide-drug conjugates to overcome the blood-brain barrier and target CNS diseases

Xue Zhou et al.

Summary: Approximately one in six people worldwide suffer from central nervous system disorders, necessitating effective strategies to improve success rates in CNS drug discovery and development. The presence of the blood-brain barrier (BBB) poses a major obstacle to CNS drug development, but brain-penetrating peptide shuttles and PDCs have shown promise in overcoming this barrier and delivering drug molecules to the brain.

Enhanced brain biodistribution of Ginsenoside Rg1 based self-nanoemulsifying drug delivery system to ameliorate metabolic syndromes and keep homeostatic balance

Abeer Khattab et al.

Summary: The study aimed to develop a self-nanoemulsifying system (SNES) based on Ginsenoside Rg1 (GRg1) to enhance its solubility, oral bioavailability, and brain targeting, while assessing its anti-obesity efficacy and safety compared to orlistat. The results showed that SNES could improve the dissolution and absorption of GRg1, reduce oxidative stress markers, suppress appetite, and prevent hepatocyte degradation. Using SNES as a carrier for GRg1 may avoid the first pass effect, increase its viability in blood, and target it to the brain in higher concentration compared to conventional extracts, making it a promising anti-obesity therapy to prevent metabolic and energy dysfunction of orlistat.

An update on oral drug delivery via intestinal lymphatic transport

Zichen Zhang et al.

Summary: Orally administered drugs must survive the gastrointestinal environment, penetrate the enteric epithelia, and avoid hepatic metabolism before reaching the systemic circulation. Hepatic metabolism is a significant barrier, and alternative pathways like intestinal lymphatic transport are being explored. Main pathways for peroral lymphatic transport include the chylomicron and microfold cell pathways, with increasing research input and applications.
