Nanoengineered Neutrophils as a Cellular Sonosensitizer for Visual Sonodynamic Therapy of Malignant Tumors

Summary: The rapid development of cell-based theranostics has attracted significant attention. In this study, customized liposomes were combined with neutrophils using nanoengineering method to generate acoustic oxygen-carrying cells called Acouscyte/O-2 for the diagnosis and treatment of cancer. Acouscyte/O-2 not only carries oxygen but also exhibits characteristics such as long circulation, inflammation-triggered recruitment and decomposition. It selectively accumulates in tumors, enhances tumor oxygen levels, and triggers anticancer sonodynamics through ultrasound stimulation, leading to complete tumor obliteration.

Combined legumain- and integrin-targeted nanobubbles for molecular ultrasound imaging of breast cancer

Summary: PD-L1 targeted therapies may be effective for various cancers. Non-invasive diagnostic and prognostic molecular imaging platforms can enhance clinical assessment of PD-L1 tumor status. By utilizing ultra-stable nanobubbles as a contrast agent, functionalized with nanobody targeting human PD-L1, molecular imaging of the tumor is possible. The results showed a three-fold increase in contrast signal compared to non-targeted nanobubbles. This research has the potential for clinical translation and prognostic evaluation of patient response to PD-L1 targeted immunotherapy.

Development of an Accurate and Robust Air-Coupled Ultrasonic Time-of-Flight Measurement Technique

Summary: This study presents a non-invasive and accurate method for in-air ranging and time-of-flight (ToF) determination that is robust to changing ambient conditions or waveform variations. It utilizes the azimuthal directivity of a laser Doppler vibrometer and autocorrelation technique to determine the in-air travel time and time of entry of the acoustic signal. The same signal is also used as a reference for determining the ToF through the specimen via cross-correlation. Experiments on a polyamide specimen verified the method, achieving a ranging accuracy of <0.1 mm and a transmission ToF accuracy of 0.3 μs.

Overcoming these barriers can help maximize and sustain the use of obstetric ultrasound technology.

Summary: Ultrasound localization microscopy has the potential to image microvasculature in vivo without sacrificing imaging penetration depth, but has been limited by the reliance on super-resolution inference. This study presents a novel anesthesia protocol in rabbits to induce safe, controllable apnea periods for long image-acquisition times. The results demonstrate the necessity of the apneic protocol for correctly identifying poorly vascularized tumor cores and revealing tumoral microvascular architecture.

Preclinical study to improve microbubble-mediated drug delivery in cancer using an ultrasonic probe with an interchangeable acoustic lens

Summary: It has been demonstrated that unfocused ultrasound with microbubbles and an interchangeable acoustic lens can enhance drug delivery efficiency by overcoming the limitations of a narrow pressure field. A concave lens suitable for an ultrasound-phased array probe was designed to optimize microbubble destruction, leading to successful drug delivery enhancement in in vitro and animal experiments. This system shows promise in providing sufficient ultrasound irradiation to microbubbles for improved drug delivery effectiveness.

Quantification of dynamic contrast-enhanced ultrasound (CEUS) in non-cystic breast lesions using external perfusion software

Summary: This clinical pilot study aimed to display typical perfusion results in patients with breast lesions using CEUS. The use of the perfusion parameters PE and AUC allowed for a good assessment of the risk of malignant breast lesions. These results provide a basis for the development of new clinical treatment methods for TNBC.
