Summary: Oxidative stress and chronic inflammation play a role in neurodegenerative diseases and inflammatory arthritis. The flavonoid-rich extract of Diplotaxis harra (DHE) showed antioxidant and anti-inflammatory activities. It inhibited iNOS and COX-2 expressions in stimulated macrophages, and the major antioxidant compound IR3G exhibited anti-inflammatory and neuroprotective activities. Molecular docking and simulation studies suggested a potential mechanism in modulating neurodegenerative diseases. This study provides evidence for the potential of D. harra-derived IR3G as a natural therapeutic agent against these diseases.

This study demonstrated that TFAP2A downregulates miR-126 and upregulates PLK2, activating the IκBα-NF-κB pathway and increasing neuronal injury following cerebral ischemia, as seen in both in-vitro and in-vivo models.

Inhibition of PLK2 shows promise in developing new therapeutics, yet the mechanism of neuroprotection through PLK2 inhibition is unlikely due to reducing α-synuclein phosphorylation. The true Lewy body kinase still awaits discovery.
