Cell cycle on the crossroad of tumorigenesis and cancer therapy

Jing Liu et al.

Summary: Cell cycle aberrancy is a fundamental mechanism underlying tumorigenesis, and therapies targeting cell cycle machinery components can not only suppress cancer cell division but also reverse cancer metabolism and restore cancer immune surveillance. Recently, proteolysis targeting chimeras (PROTACs) have been used to target oncogenic proteins related to cell cycle progression for more effective tumor suppression.

Elevated METTL9 is associated with peritoneal dissemination in human scirrhous gastric cancers

Toshifumi Hara et al.

Summary: Methyltransferase-like 9 (METTL9) plays a crucial role in the metastasis of scirrhous gastric cancer. Its elevated expression is closely associated with enhanced metastatic activity. Knockdown of METTL9 significantly inhibits migration and invasion of metastatic cells. METTL9 is predominantly localized in mitochondria in metastatic cells, and its knockdown reduces mitochondrial Complex I activity. Therefore, METTL9 may be a potential molecular target to inhibit peritoneal dissemination of scirrhous gastric cancer.

Epigenetic Biomarkers for the Diagnosis and Treatment of Liver Disease

Maria Arechederra et al.

Summary: Cellular identity and function are regulated by the correct expression of genetic information, which is controlled by various epigenetic mechanisms such as DNA methylation, histone modifications, and non-coding RNA expression. Abnormalities in these mechanisms are associated with the development of multiple diseases. Chronic liver disease, including hepatocellular carcinoma, is on the rise globally, but management of these patients is challenging due to late diagnosis and lack of effective therapies. Understanding and targeting epigenetic alterations may provide new therapeutic strategies and diagnostic tools for these diseases.

The language of chromatin modification in human cancers

Shuai Zhao et al.

Summary: The deregulation of chromatin modification underlies numerous oncogenic processes, contributing to cancer development by inducing epigenetic, transcriptomic, and phenotypic alterations. Understanding and developing more effective drugs to target these chromatin-related vulnerabilities will enhance the treatment of human malignancies. This review synthesizes the connections between chromatin modifications and cancer, highlighting options for therapeutic targeting based on recent advances.
