The Smad4-MYO18A-PP1A complex regulates beta-catenin phosphorylation and pemigatinib resistance by inhibiting PAK1 in cholangiocarcinoma

Jialiang Liu et al.

Summary: This study revealed that coexpression of Smad4 and MYO18A is a favorable prognostic indicator for iCCA and pCCA, suppressing CCA proliferation and metastasis by regulating beta-catenin phosphorylation and intracellular localization.

HJURP Promotes Malignant Progression and Mediates Sensitivity to Cisplatin and WEE1-inhibitor in Serous Ovarian Cancer

Zhiyuan Dou et al.

Summary: This study found that HJURP is overexpressed in ovarian cancer and is associated with unfavorable prognosis. Knockdown of HJURP inhibits proliferation, metastasis, and induces cell stagnation in ovarian cancer cells. Mechanistically, HJURP regulates WEE1 through the MYC pathway, affecting chemosensitivity and DNA repair in ovarian cancer cells. Moreover, silencing HJURP enhances sensitivity to cisplatin and improves the synergistic effect of combined therapy with cisplatin and AZD1775 in ovarian cancer cells.

Wnt-TCF7-SOX9 axis promotes cholangiocarcinoma proliferation and pemigatinib resistance in a FGF7-FGFR2 autocrine pathway

Zengli Liu et al.

Summary: Cholangiocarcinoma (CCA) is a highly malignant tumor originating from bile ducts. This study identified SOX9 and FGF7 as prognostic biomarkers of CCA and demonstrated that the WNT3A-TCF7-SOX9 axis can induce pemigatinib resistance through two independent pathways.

HJURP regulates cell proliferation and chemo-resistance via YAP1/NDRG1 transcriptional axis in triple-negative breast cancer

Misha Mao et al.

Summary: This study focuses on the important role of the HJURP/YAP1/NDRG1 transcriptional regulation axis in triple-negative breast cancer. It found that HJURP affects the transcriptional activity of YAP1 and regulates cell proliferation and chemotherapy sensitivity by modulating the transcription of NDRG1 in triple-negative breast cancer.

HMGA1-TRIP13 axis promotes stemness and epithelial mesenchymal transition of perihilar cholangiocarcinoma in a positive feedback loop dependent on c-Myc

Zhipeng Li et al.

Summary: This study identified two potential unfavorable prognostic biomarkers, HMGA1 and TRIP13, for pCCA, and revealed their roles in promoting progression through enhancing stemness, epithelial-mesenchymal-transition, proliferation, migration, and invasion. Additionally, the HMGA-TRIP13 axis also strengthened EMT and stemness through a positive feedback pathway dependent on c-Myc.

WDR5 facilitates EMT and metastasis of CCA by increasing HIF-1α accumulation in Myc-dependent and independent pathways

Tianli Chen et al.

Summary: This study demonstrates that WDR5 interacts with c-Myc in cholangiocarcinoma (CCA), leading to increased HIF-1 alpha expression and stability, thereby promoting tumor metastasis and invasion.

FGF19 and FGFR4 promotes the progression of gallbladder carcinoma in an autocrine pathway dependent on GPBAR1-cAMP-EGR1 axis

Tianli Chen et al.

Summary: The study revealed that FGF19 and FGFR4 are adverse prognostic biomarkers for gallbladder carcinoma, with their co-expression being a more sensitive predictor. FGF19 can promote GBC progression by stimulating FGFR4 activation via an autocrine pathway with bile as a potential carrier.

HJURP promotes proliferation in prostate cancer cells through increasing CDKN1A degradation via the GSK3β/JNK signaling pathway

Wenjie Lai et al.

Summary: Genes with cross-cancer aberrations, such as HJURP, play a potential role in cancer progression and could serve as therapeutic targets. Higher expression of HJURP is associated with poorer outcomes in prostate cancer patients, and its mechanism involves promoting cell proliferation by regulating CDKN1A ubiquitination levels.
