Future perspectives of uveal melanoma blood based biomarkers

Aaron B. Beasley et al.

Summary: Uveal melanoma is the most common primary intraocular malignancy, and despite successful local treatment, the risk of metastatic disease remains. Intraocular tumor biopsy is the current prognostic method, but it is invasive and limited by sampling variability. Liquid biopsy, including circulating tumor cells, circulating tumor DNA, microRNA, and exosomes, holds great potential for minimally invasive genetic characterization of tumors.

Overexpression of microRNA-130a represses uveal melanoma cell migration and invasion through inactivation of the Wnt/β-catenin signaling pathway by downregulating USP6

Shuai Wu et al.

Summary: Uveal melanoma (UM) is the most common primary malignant intraocular tumor, with microRNA-130a (miR-130a) underexpressed in UM patients. Low expression of miR-130a in UM tissues, especially in metastatic tissues, correlates with shorter overall survival. Overexpression of miR-130a inhibits USP6 expression, suppressing UM cell migration and invasion abilities through inactivating the Wnt/beta-catenin signaling pathway.

Uveal melanoma pathobiology: Metastasis to the liver

Prisca Bustamante et al.

Summary: Uveal melanoma is an intraocular tumor that tends to metastasize to the liver. Despite identifying early driver mutations, the process of UM metastasis remains incompletely understood. A better understanding of genetic, molecular, and environmental factors could lead to new approaches for UM treatment.

Co-deletion of Lrp5 and Lrp6 in the skeleton severely diminishes bone gain from sclerostin antibody administration

Kyung-Eun Lim et al.

Summary: In conclusion, unlike with the deletion of Lrp5 alone, the bone-selective late-stage co-deletion of Lrp5 and Lrp6 significantly impairs or completely nullifies the osteogenic action of Scl-mAb, and highlights a major role for both Lrp5 and Lrp6 in the mechanism of action for the bone-building effects of sclerostin antibody.

Single-cell RNA sequencing reveals intratumoral heterogeneity in primary uveal melanomas and identifies HES6 as a driver of the metastatic disease

Charlotte Pandiani et al.

Summary: The study used single-cell RNA sequencing to uncover intratumoral heterogeneity in primary uveal melanomas, identified a gene regulatory network related to invasive and poor prognosis states, validated the role of transcription factor HES6 in tumors, and demonstrated the essential role of HES6 in growth and metastasis of uveal melanomas.

Piezo type mechanosensitive ion channel component 1 facilitates gastric cancer omentum metastasis

Xiaofei Wang et al.

Summary: The study revealed that Piezo1 plays a crucial role in gastric cancer omentum metastasis by promoting cell proliferation and migration, impacting intracellular Ca2+ influx through upregulation of HIF-1α, hence affecting tumor growth and metastasis.

Bone-derived sclerostin and Wnt/β-catenin signaling regulate PDGFRα+ adipoprogenitor cell differentiation

Soohyun P. Kim et al.

Summary: Sclerostin, an antagonist of the Wnt signaling pathway, regulates the communication between bone and adipose tissue, influencing adipocyte formation and metabolism through various mechanisms. Studies have shown that deficiency of sclerostin can lead to an increase in adipocyte progenitors, a decrease in fat mass, and improved glucose metabolism.

In situ immunogenic clearance induced by a combination of photodynamic therapy and rho-kinase inhibition sensitizes immune checkpoint blockade response to elicit systemic antitumor immunity against intraocular melanoma and its metastasis

Seohyun Kim et al.

Summary: A targeted immunotherapeutic strategy involving local photodynamic therapy, a rho-kinase inhibitor, and PD-1/PD-L1 immune checkpoint blockade was proposed for uveal melanoma. This treatment approach demonstrated efficacy in promoting immunogenic clearance of primary tumors, preventing metastasis, and improving survival outcomes for patients.

Proteomics of Primary Uveal Melanoma: Insights into Metastasis and Protein Biomarkers

Geeng-Fu Jang et al.

Summary: This study conducted proteomic analysis of primary uveal melanoma (pUM) to investigate metastasis mechanisms and protein biomarkers. The findings revealed 402 differentially expressed proteins between metastasizing and non-metastasizing pUM, suggesting significant differences in the immune response. Predictive modeling identified 32 proteins capable of distinguishing metastasizing from non-metastasizing pUM with 93% accuracy, supporting the potential for protein-based prognostic methods for detecting uveal melanoma metastasis.

Immune Checkpoint Blockade for Metastatic Uveal Melanoma: Patterns of Response and Survival According to the Presence of Hepatic and Extrahepatic Metastasis

Elias A. T. Koch et al.

Summary: This study found that patients with metastatic uveal melanoma showed a better response to dual immune checkpoint blockade when there were extrahepatic metastases present. Both hepatic and extrahepatic metastases were associated with improved survival and treatment response compared to hepatic metastases alone.

ZFPM2-AS1 transcriptionally mediated by STAT1 regulates thyroid cancer cell growth, migration and invasion via miR-515-5p/TUSC3

Ruizhen Ren et al.

Summary: In thyroid cancer, ZFPM2-AS1 acts as an oncogene mediated by STAT1, weakening the inhibitory effect of miR-515-5p on TUSC3 and promoting cancer progression. Thus, ZFPM2-AS1 may serve as a potential biomarker for thyroid cancer.
