Identification and quantification of immune infiltration landscape on therapy and prognosis in left- and right-sided colon cancer

Jun-Nan Guo et al.

Summary: The study aimed to identify immune cell infiltration (ICI) subtypes for evaluating prognosis and therapeutic benefits in left-sided and right-sided colon cancer. High ICI scores were associated with increased tumor mutation burden (TMB) and genomic instability related signaling pathways. The study found significant differences in prognosis, sensitivities to targeted inhibitors and immunotherapy, IPS, and expression of immune checkpoints between high and low ICI score groups, with key genes including CA2 and TSPAN1 identified by ICI scores at various levels.

Conjugate of ibrutinib with a TLR7 agonist suppresses melanoma progression and enhances antitumor immunity

Sumei Ren et al.

Summary: The use of large molecules in immunotherapy has shown promising developments in cancer treatment. However, small molecule targeted therapies still lack effective immune-functional classes. In this study, researchers developed a strategy to create novel small molecule compounds that can target tumors and stimulate the immune system. The compound GY161 demonstrated positive effects in stimulating immune response and inhibiting melanoma cell growth in vitro and in vivo. These findings suggest that this strategy could be a reliable and promising approach for developing small molecule immunogenic anticancer drugs.

NLRC3 High Expression Represents a Novel Predictor for Positive Overall Survival Correlated With CCL5 and CXCL9 in HCC Patients

Chengpan Wang et al.

Summary: NLRC3 has been reported as a factor inhibiting inflammatory responses. Its role in hepatocellular carcinoma (HCC) is still unknown. In this study, NLRC3 levels were evaluated using the GEO database and multiple immunohistochemical analysis with tumor tissue microarrays of HCC patients. Tumor-bearing mouse models were also established to assess the effect of NLRC3. The results showed that high NLRC3 expression was associated with favorable overall and disease-free survival. NLRC3 was also found to be involved in CD8(+) T cell infiltration and correlated with increased CCL5 and CXCL9 expressions in the tumor microenvironment. These findings suggest that boosting NLRC3 could be a promising treatment to enhance survival in HCC patients.

Memory-like Differentiation Enhances NK Cell Responses to Melanoma

Summary: The treatment of advanced melanoma is a clinical challenge, and natural killer (NK) cells show promise as a cellular therapy. Cytokine-induced memory-like (ML) differentiation enhances NK-cell responses against blood cancer targets, but its activity against solid tumors like melanoma needs further research. In preclinical studies, ML NK cells from healthy donors and melanoma patients exhibited increased IFN gamma production and cytotoxicity against melanoma targets, with a response partially dependent on activating receptors NKG2D and NKp46. Furthermore, in mouse models, ML NK cells showed better control of melanoma compared to conventional NK cells.
