High APOBEC1 Complementation Factor Expression Positively Modulates the Proliferation, Invasion, and Migration of Endometrial Cancer Cells Through Regulating P53/P21 Signaling Pathway

Qin Liu et al.

Summary: This study found that A1CF is highly expressed in endometrial cancer (EC) and affects the proliferation, invasion, and migration of EC cells through the regulation of the P53/P21 signaling pathway. High A1CF expression is associated with poor prognosis in EC, suggesting it as a potential therapeutic target.

Lipid reprogramming induced by the TFEB-ERRα axis enhanced membrane fluidity to promote EC progression

Xiaodan Mao et al.

Summary: This study reveals that lipid reprogramming induced by the TFEB-ERRα axis enhances the progression of endometrial cancer through EMT signaling. The TFEB-ERRα axis increases membrane fluidity, promotes pseudopod formation, and enhances cell invasion. TFEB and ERRα are highly expressed in EC patients and correlated with EC progression. Furthermore, the TFEB-ERRα axis affects lipid metabolism by altering mitochondrial function.

Long-term quality of life outcomes of women treated for early-stage endometrial cancer

Saira Sanjida et al.

Summary: The study compared long-term quality of life in women treated for early-stage endometrial cancer with population norms and compared quality of life outcomes of patients receiving different surgical treatments. Results showed that long-term endometrial cancer survivors reported higher prevalence of anxiety/depression across all age groups, and deficits in mobility and usual activities for women aged >= 75 years. Physical and functional well-being were better among women who underwent total laparoscopic hysterectomy compared to total abdominal hysterectomy recipients.

Mitochondrial oxidative metabolism contributes to a cancer stem cell phenotype in cholangiocarcinoma

Chiara Raggi et al.

Summary: The study reveals that cancer stem cells in cholangiocarcinoma have a more efficient respiratory phenotype, relying on mitochondrial oxidative metabolism and PGC-1a to maintain CSC features. Targeting mitochondrial complex I or PGC-1a in CCA-SPH impairs spherogenicity and expression of markers related to CSC phenotype, pluripotency, and epithelial-mesenchymal transition. Administration of metformin or SR-18292 in mice with CCA-SPH xenografts significantly reduces tumor growth and reverts the phenotype to that of cells grown in monolayer.

Acriflavine, a Potent Inhibitor of HIF-1α, Disturbs Glucose Metabolism and Suppresses ATF4-Protective Pathways in Melanoma under Non-hypoxic Conditions

Roman Marti-Diaz et al.

Summary: The study found that HIF-1α is constitutively expressed in melanoma cells under normoxic conditions and its elevated expression is associated with the aggressiveness of melanoma tumors. Using acriflavine as an inhibitor of HIF-1α dimerization, the researchers investigated whether HIF-1α-regulated pathways contribute to the growth of melanoma cells under normoxia. Results showed that acriflavine can differentially modulate HIF-1α-regulated targets, inhibiting glucose metabolism and the Warburg effect, ultimately leading to cell death.

Inhibition of Estrogen-Related Receptor α Blocks Liver Steatosis and Steatohepatitis and Attenuates Triglyceride Biosynthesis

Chien-Yu Chen et al.

Summary: Inhibiting ERR alpha can block the development of NAFLD, reduce lipid accumulation, and attenuate NASH. Glycerolipid synthesis was identified as an additional mechanism for ERR alpha-regulated NAFLD/NASH development, with glycerophosphate acyltransferase 4 identified as a novel transcriptional target of ERR alpha. These results establish ERR alpha as a potential target for NAFLD/NASH treatment and highlight novel signaling pathways regulated by ERR alpha.

Deciphering the Pyroptosis-Related Prognostic Signature and Immune Cell Infiltration Characteristics of Colon Cancer

Ran Wei et al.

Summary: This study identified 27 pyroptosis-related regulators and 854 signature genes associated with them, establishing an eight-gene assessment model for predicting prognosis in colon cancer patients. Three distinct pyroptosis-related molecular patterns were correlated with different clinical outcomes and immune cell infiltration characteristics in the tumor microenvironment. The study suggests that understanding pyroptosis-related patterns and models can provide insights into immune cell infiltration characteristics in the tumor microenvironment and guide future research on targeting pyroptosis in cancer.
