Lactic acid promotes PD-1 expression in regulatory T cells in highly glycolytic tumor microenvironments

Summary: In highly glycolytic tumors, regulatory T (Treg) cells have higher PD-1 expression compared to effector T cells. Under low-glucose conditions, Treg cells actively absorb lactic acid through monocarboxylate transporter 1 (MCT1), promoting the expression of PD-1, while PD-1 expression by effector T cells is dampened. PD-1 blockade invigorates PD-1-expressing Treg cells, resulting in treatment failure.

High levels of serum succinate have been found in patients and mouse models with cardiovascular disease. Atherosclerosis is the pathological basis of cardiovascular and peripheral vascular diseases, affecting human health significantly.

Summary: This study explores the interplay between sphingolipid metabolism and mitochondrial bioenergetics in cancer cells and suggests that targeting both pathways could be a novel treatment strategy against multidrug resistance. Deficiencies in sphingolipid metabolism and the oxidative phosphorylation system are observed in multidrug resistant leukemia cells, and a combinatorial therapeutic approach targeting both pathways induces synergistic cytotoxicity.

Summary: Unlike most cell types, cancer cells can survive in low acid environments. Knocking out genes associated with pH-related fitness defects in colorectal cancer cells can cause cell death under acidic conditions. Researchers have also discovered mitochondrial metabolism inhibitors that can specifically kill cancer cells in a low acid environment. The growth of xenografts established from gene-knockout cells is slower, but can be stimulated with bicarbonate therapy to reduce tumor acid stress.

Long Term Survivals in Aggressive Primary Brain Malignancies Treated With an Adjuvant Ketogenic Diet

Summary: Aggressive primary brain tumors such as glioblastoma multiforme and grade IV astrocytoma are typically treated with surgery, radiation therapy, and chemotherapy. The addition of a ketogenic diet to rodents with brain tumors has shown promising results in increasing survival. In this study, adult patients with APBT underwent neurosurgery followed by an adjuvant ketogenic diet concurrent with radiation therapy and temozolomide. Younger patients showed longer survival rates compared to older patients. Further research is needed to confirm these findings, particularly in younger patients under the age of 40.

Summary: This case report describes a 32-year-old male patient with IDH1-mutant GBM who was successfully treated with ketogenic metabolic therapy and surgical debulking. The patient has maintained a good quality of life with long-term survival, potentially due to a therapeutic metabolic synergy between KMT and the IDH1 mutation. Further research is needed to determine the effectiveness of this non-toxic therapeutic strategy for other GBM patients with or without IDH mutations.

Patterns of glioblastoma treatment and survival over a 16-years period: pooled data from the German Cancer Registries

Summary: This study analyzed 40,138 adult GBM cases diagnosed in Germany between 1999 and 2014, finding that younger age, female sex, surgical resection, and the use of radiotherapy and chemotherapy were associated with better survival outcomes. Survival improvements were observed over time for each treatment option.
