Wnt and PI3K/Akt/mTOR Survival Pathways as Therapeutic Targets in Glioblastoma

Amir Barzegar Behrooz et al.

Summary: Glioblastoma (GBM) is a devastating type of brain tumor, and targeting the Wnt/β-catenin and PI3K/Akt/mTOR (PAM) signaling pathways, which play crucial roles in GBM progression and invasion, may provide important advancements in alternative treatment strategies for GBM.

RNASE2 Mediates Age-Associated B Cell Expansion Through Monocyte Derived IL-10 in Patients With Systemic Lupus Erythematosus

Yantong Zhu et al.

Summary: Systemic lupus erythematosus (SLE) is characterized by the production of pathogenic autoantibodies. This study found that Ribonuclease A family member 2 (RNase2) is highly expressed in SLE patients and is associated with the increase of autoreactive B cells known as age-associated B cells (ABCs). Reduction of RNASE2 expression led to a decrease in ABCs, accompanied by a reduction in total IgG and IL-10 levels. It was also found that both RNASE2 and IL-10 in SLE patients were mainly derived from monocytes. The study concluded that RNASE2 might induce the production of ABCs via IL-10 secreted from monocytes, contributing to the pathogenesis of SLE.

Selective cleavage of ncRNA and antiviral activity by RNase2/EDN in THP1-induced macrophages

Lu Lu et al.

Summary: RNase2, the most abundant member of the RNaseA family in macrophages, has been found to have antiviral effects. In this study, RNase2 was knocked out in THP-1 cells and the response to Respiratory Syncytial Virus (RSV) was analyzed. RSV was found to induce expression of RNase2, which significantly enhanced cell survival. Sequencing analysis revealed that tRNA-derived fragments (tRFs) and second miRNAs were mostly accumulated in WT cells compared to KO cells.

Identification of a novel immune prognostic model in gastric cancer

Y. Li et al.

Summary: This study developed an immune-related risk model for predicting prognosis and assessing the immune status of gastric cancer patients. The model, based on seven immune-related genes, showed potential in stratifying patients into different risk groups with different prognostic outcomes. Furthermore, the high-risk group exhibited higher levels of immunosuppressive cells and inhibitory immune checkpoints, suggesting a suppressive immune microenvironment in these patients.

Identification of Potential Biomarkers and Immune Infiltration Characteristics in Idiopathic Pulmonary Arterial Hypertension Using Bioinformatics Analysis

Haowei Zeng et al.

Summary: HBB, RNASE2, S100A9, and IL1R2 were identified as potential biomarkers to discriminate IPAH from the control group. There was a significant difference in immune infiltration between patients with IPAH and the normal group, with higher levels of certain immune cells observed in IPAH patients.

Summary: This review of recent clinical trials on targeted therapies against the PI3K-AKT-mTOR pathway found that pan-PI3K inhibitors and selective PI3K inhibitors have not shown significant clinical efficacy. AKT inhibitors may have significant adverse effects, but showed some evidence of improved survival.
