NLRP3 Inflammasome Activation Controls Vascular Smooth Muscle Cells Phenotypic Switch in Atherosclerosis

Summary: The study demonstrated that oxLDL-activated monocytes can directly affect VSMCs in a co-culture system, leading to reduced expression of certain markers and upregulation of others, as well as activation of caspase 1, secretion of IL-1 beta, and pyroptosis in VSMCs. The activation of VSMC NLRP3 inflammasome by monocytes may play a detrimental role in atherosclerotic plaque stability in human atherosclerosis, as evidenced by findings in both mice and human atherosclerotic plaques.

Summary: CD8(+)T cells play a crucial role in tumor elimination, and Peli1 is a protein associated with innate immunity. This study found that increased expression of Peli1 is associated with an increased risk of developing tumors and a decrease in the effector function of CD8(+)T cells. On the other hand, a deficiency in Peli1 enhances the activity of CD8(+)T cells. Additionally, Peli1 interacts with PKC-θ and inhibits its downstream signaling pathway in T cells.

These macrophages promote the transdifferentiation of VSMCs into osteogenic-like cells through the secretion of CCL24. The abundance of LYVE-1(+)CCL24(+) macrophages and elevated systemic levels of CCL24 are associated with vascular calcification and CAD events.

The Role of Ubiquitin E3 Ligase in Atherosclerosis

Summary: Atherosclerosis is a chronic inflammatory vascular disease that is the main cause of death in both developed and developing countries. Evidence suggests that ubiquitination plays important roles in the pathogenesis of atherosclerosis, including regulation of vascular inflammation, endothelial cell and vascular smooth muscle cell function, lipid metabolism, and plaque stability. Targeting ubiquitin E3 ligases may provide a novel strategy for preventing the progression of atherosclerosis.

Summary: Inflammasomes are important for innate immunity against infections but can also contribute to inflammatory diseases when deregulated. Peli1, as an E3 ubiquitin ligase, plays a critical role in regulating the activation of NLRP3 inflammasome by promoting ASC ubiquitination and facilitating inflammasome assembly. Deficiency in Peli1 impairs NLRP3-induced caspase-1 activation and IL-1 beta maturation, highlighting its significance as an inflammasome regulator.
