Macrophages play a crucial role in confining the dissemination of Mtb, but the virulence factors of Mtb can disrupt macrophage homeostasis, leading to active TB, especially during suppressed or hyperactivated immune states.

The results showed that patients with T2DM had significantly increased levels of Th17 cells, IL-17 and IL-10, but decreased levels of Treg cells and Treg/Th17 ratio compared to the controls.

FAK plays an important role in host defense against Mtb infection by preventing necrotic cell death and inducing ROS production to restrict bacterial survival.

Alveolar Macrophages: Adaptation to Their Anatomic Niche during and after Inflammation

Summary: Alveolar macrophages (AM) are colonized by embryonic macrophages in early lung development and develop into resident alveolar macrophages (ResAM), whose functions and programming are influenced by the lung microenvironment. Infections deplete the pool of ResAM, allowing for the recruitment of inflammatory monocyte-derived alveolar macrophages (InfAM) to develop and eventually gain profibrotic functions for tissue healing. The memory of these cells, called trained immunity, can have both advantages in defense capacities and drawbacks in perpetuating harmful processes.

Dapagliflozin reverses the imbalance of T helper 17 and T regulatory cells by inhibiting SGK1 in a mouse model of diabetic kidney disease

Summary: An imbalance between T helper 17 (Th17) and T regulatory (Treg) cell subsets contributes to the pathogenesis of diabetic kidney disease (DKD). This study demonstrated that dapagliflozin (Dap) can inhibit SGK1 to suppress the polarization of Th17/Treg cells in diabetic kidneys, leading to attenuation of albuminuria and tubulointerstitial fibrosis independent of glycemic control. These results suggest that the imbalance of Th17/Treg cells plays an important role in DKD and Dap protects against DKD by reversing the T-cell imbalance.
