The multifaceted role of ferroptosis in liver disease

Junyi Chen et al.

Summary: Ferroptosis is an iron-dependent form of non-apoptotic cell death characterized by excessive lipid peroxidation and is associated with liver diseases. Dysregulated metabolic pathways and impaired iron homeostasis play a role in the progression of liver disease via ferroptosis. Several ferroptosis-associated genes and pathways have been implicated in liver disease. Targeting ferroptosis may have therapeutic potential for managing liver diseases.

PKCβII phosphorylates ACSL4 to amplify lipid peroxidation to induce ferroptosis

Clinical use of N-acetyl cysteine during liver transplantation: Implications of oxidative stress and inflammation as therapeutic targets

Yonela Ntamo et al.

Summary: Liver transplantation is the definitive treatment for complete liver failure, but oxidative stress and inflammation contribute to the damage caused during the procedure. N-acetyl cysteine (NAC) has been studied as a potential therapeutic drug to counteract oxidative stress and inflammation. Clinical evidence suggests that NAC can improve liver function by enhancing endogenous antioxidants and attenuating inflammation, but some studies have shown conflicting results.

Epidemiology of cardiovascular disease in Europe

Nick Townsend et al.

Summary: This review highlights the significant health burden of cardiovascular disease (CVD) in the WHO European Region, with CVD being the leading cause of death in the region. Men have higher age-standardized rates of morbidity and death from CVD compared to women, particularly in individuals under 70 years of age. Improving surveillance and monitoring, as well as implementing evidence-based preventive and treatment approaches, are essential to address the large inequalities in CVD outcomes across countries in Europe.

Vitamin K1 inhibits ferroptosis and counteracts a detrimental effect of phenprocoumon in experimental acute kidney injury

Benedikt Kolbrink et al.

Summary: Ferroptosis, a type of iron-dependent programmed cell death, plays a vital role in multiple diseases. However, there are no pharmacologic inhibitors of ferroptosis in clinical use. In this study, vitamin K1 was identified as an efficient inhibitor of ferroptosis, providing a new strategy for pharmacological control of this mode of cell death.

Ferroptotic pores induce Ca2+ fluxes and ESCRT-III activation to modulate cell death kinetics

Lohans Pedrera et al.

Summary: Ferroptosis is a form of regulated necrosis that depends on iron ions and is associated with lipid peroxidation. An increase in cytosolic Ca2+ precedes cell bursting in ferroptosis, and membrane nanopores are linked to plasma membrane damage in this process. Activation of ESCRT-III serves as a general protective mechanism, counterbalancing cell death kinetics during ferroptosis.

Dopaminergic Therapy for Motor Symptoms in Early Parkinson Disease Practice Guideline Summary A Report of the AAN Guideline Subcommittee

Summary: The review found that initial treatment with levodopa provides better motor benefits compared to dopamine agonists, but is also more likely to cause motor complications. There was little evidence to suggest that any particular formulation or method of administration of dopamine agonists is superior. Long-acting forms of levodopa and combination formulations with entacapone do not appear to differ in efficacy from immediate-release levodopa for early disease motor symptoms. Patients using dopamine agonists may have a higher risk of impulse control disorders compared to those using levodopa.

N-Acetylcysteine for Cardiac Protection During Coronary Artery Reperfusion: A Systematic Review and Meta-Analysis of Randomized Controlled Trials

Sher Ali Khan et al.

Summary: Reperfusion is essential for managing symptoms in patients with myocardial ischemia, but it can lead to paradoxical injury. The use of N-acetylcysteine (NAC) has shown promise in reducing reactive oxygen species (ROS) and improving clinical outcomes in patients undergoing coronary artery reperfusion procedures. Meta-analysis of randomized controlled trials (RCTs) suggests that NAC administration during or prior to reperfusion may decrease serum cardiac troponin levels and the incidence of postoperative atrial fibrillation, providing supportive evidence for the potential benefit of NAC as adjunct therapy for cardiac protection against reperfusion injury.
