Iron-Chelation Treatment by Novel Thiosemicarbazone Targets Major Signaling Pathways in Neuroblastoma

Peter Macsek et al.

Summary: Novel thiosemicarbazones show promise in targeting MYCN-amplified and non-amplified neuroblastoma by reducing N-MYC and c-MYC expression, and decreasing total EGFR levels and phosphorylation through the involvement of NDRG1, a positive prognostic marker in neuroblastoma.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Synergistic efficacy of inhibiting MYCN and mTOR signaling against neuroblastoma

Matthew J. Kling et al.

Summary: The study demonstrated the potential therapeutic efficacy of targeting dysregulated protein synthesis pathways by inhibiting MYCN and mTOR signaling simultaneously in NB, particularly showing significant effects on MYCN-amplified NB cells. Further preclinical evaluation is needed to assess the clinical utility of this targeted therapy for high-risk NB patients.

Molecular Mechanisms of MYCN Dysregulation in Cancers

Ruochen Liu et al.

Summary: MYCN, a member of the MYC proto-oncogene family, encodes the transcription factor N-MYC. Abnormal expression of N-MYC is associated with high-risk cancers and poor prognosis. Therapeutic approaches for targeting MYCN-driven tumors focus on disrupting transcription, translation, protein stability, and synthetic lethality of MYCN.

Targeting Mitochondrial Metabolism in Clear Cell Carcinoma of the Ovaries

Xiaonan Zhang et al.

Summary: Ovarian clear cell carcinoma (OCCC) is a rare and chemorefractory tumor with about 50% of patients having inactivating mutations in the ARID1A gene. Loss of ARID1A leads to increased mitochondrial metabolism in OCCC cells, making them more dependent on mitochondrial activity. Preclinical testing has shown that targeting mitochondrial activity can extend overall survival in ARID1A-mutated OCCC.

A small-molecule pan-class I glucose transporter inhibitor reduces cancer cell proliferation in vitro and tumor growth in vivo by targeting glucose-based metabolism

Pratik Shriwas et al.

Summary: DRB18 is a potent pan-class I GLUT inhibitor that reduces tumor growth by inhibiting glucose transport and metabolism mediated by GLUT1-4. The inhibition of pan-class I GLUT is more effective in inhibiting tumor growth compared to single GLUT targeting.
