Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

A meta-analysis of anal cancer incidence by risk group: Toward a unified anal cancer risk scale

Gary M. Clifford et al.

Summary: Certain population groups such as PLHIV, MSM, SOTRs, and patients with autoimmune diseases have higher anal cancer risks. The incidence rates vary significantly among different groups, with HIV-positive MSM showing the highest rates. Prioritizing anal cancer prevention and research in these high-risk groups is crucial.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

An update on COVID-19 pandemic: the epidemiology, pathogenesis, prevention and treatment strategies

Hin Fung Tsang et al.

Summary: The transmission of COVID-19 remains uncontrollable, with increasing numbers of confirmed and death cases reported worldwide. There is currently no specific antiviral treatment for COVID-19, and treatment guidelines vary between countries. However, combination therapies and repurposing FDA-approved drugs show promise in clinical improvement for severe COVID-19 patients. Generating an ecosystem with effective protection, precautions, and preventive measures is crucial in the absence of specific treatment and vaccines for COVID-19.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The development of the human vaginal fornix and the portiocervicis

Helga Fritsch et al.

Summary: Our study on prenatal human development reveals the presence of two different cell lineages meeting at the transitional zone of the cervix uteri, with differentiation depending on signals from the stromal compartment. As development progresses, the epithelia and stroma of the cervical canal gradually mature in fetuses, with initial functional differentiation observed in the perinatal stages.

Summary: This systematic review suggests that women with HPV- and p16-positive vaginal cancer have a better prognosis compared to those with HPV- or p16-negative vaginal cancer. Results for p53 were varied, and no definitive conclusion could be drawn. Further larger studies are needed to determine the prognostic impact of HPV, p16, and p53 in vaginal cancer.

Matrix Metalloproteinases and Tissue Inhibitors of Metalloproteinases in Echinoderms: Structure and Possible Functions

Igor Yu Dolmatov et al.

Summary: Echinoderms, an ancient group of invertebrates, have a wide variety of matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) in their genomes. Phylogenetic analysis shows that MMPs and TIMPs underwent duplication and divergence after the separation of Echinoderms and Chordata. The MMPs of echinoderms have lytic activity towards collagen type I and gelatin, playing important roles in development, asexual reproduction, and regeneration.

Summary: The study aimed to provide an updated overview of regression, persistence, and progression rates of conservatively managed CIN 1/CIN 2/CIN 3. Most CIN 1/CIN 2 cases will regress spontaneously in less than 24 months, with the highest regression rates seen in high-risk HPV-negative and young women, while progression to cancer is less than 0.5%. Conservative management may be considered, especially in fertile women with expected high compliance.
