A Recent Ten-Year Perspective: Bile Acid Metabolism and Signaling

Yulia Shulpekova et al.

Summary: Bile acids play crucial physiological roles in the human body. They are essential for the absorption and excretion of nutrients and also act as sensors for intestinal contents, regulating their effects through processes such as intestinal absorption. Bile acids have significant impacts on liver function, cholesterol synthesis, and detoxification of xenobiotics. Their synthesis and regulation in different parts of the digestive tract are vital, as is their interaction with the intestinal microbiota.

Bile Acids: Key Players in Inflammatory Bowel Diseases?

Aicha Kriaa et al.

Summary: Inflammatory bowel diseases (IBDs) are a global public health problem with limited effective therapeutic options. Changes in gut microbiota composition and alterations in bile acids (BAs) have been identified as important factors in dysregulated intestinal immunity and the pathogenesis of IBDs. This review explores the interactions between BAs and gut microbiota in IBDs and discusses how the gut microbiota-BA-host axis may influence digestive inflammation.

Association of mast cell density, microvascular density and endothelial area with clinicopathological parameters and prognosis in canine mammary gland carcinomas

Simona Sakalauskaite et al.

Summary: This study investigated the association between mast cell density and clinicopathological parameters in canine mammary tumors. The results showed that intratumoural mast cell density was associated with tumor size, lymph node involvement, and tumor-infiltrating lymphocyte count, while peritumoural mast cell density was associated with tumor grade. The study also found that high-grade tumors with high total intratumoural mast cell density and a larger endothelial area were associated with shorter disease-free survival. In addition, intratumoural mast cell density and grade were identified as independent prognostic factors.

KHDRBS3 promotes multi-drug resistance and anchorage-independent growth in colorectal cancer

Shoichi Ukai et al.

Summary: The study focused on the KHDRBS3 gene and its potential role in colorectal cancer drug resistance. KHDRBS3 was found to be associated with poor prognosis in CRC patients and may regulate stem cell properties through CD44 variant expression and the Wnt signaling pathway. KHDRBS3 could serve as a promising candidate marker for predicting chemotherapy outcomes and prognosis in CRC patients.

Interference of S100A16 suppresses lipid accumulation and inflammation in high glucose-induced HK-2 cells

Lanlan Liu et al.

Summary: The study found that S100A16 knockdown could protect against high glucose-induced lipid accumulation and inflammation in HK-2 cells through regulating SCAP/SREBP1 signaling. Lipid droplets and cholesterol were reduced, as well as the production of inflammatory cytokines. Additionally, S100A16 knockdown decreased the expression of SCAP, SREBP1, SCD1, and SCAP, but the inhibitory effect was reversed by SREBP1 overexpression.

Salivary CPLANE1 Levels as a Biomarker of Oral Squamous Cell Carcinoma

Sei Ueda et al.

Summary: The study identified salivary CPLANE1 as a potential biomarker for screening and early detection of oral squamous cell carcinoma (OSCC). CPLANE1 expression levels were significantly higher in OSCC patients compared to healthy volunteers and OPMDs patients. A screening test utilizing CPLANE1 showed good accuracy for detecting OSCC.

TFF1 Induces Aggregation and Reduces Motility of Helicobacter pylori

Daniela Eletto et al.

Summary: H. pylori infection is a major risk factor for gastric cancer development, with TFF1 playing a protective role by affecting the aggregation and motility of the bacterium, as well as reducing gene transcription. This interaction may explain the persistent presence of H. pylori in the human host without causing disease.

Valerio Baldelli et al.

Summary: This article discusses the significant contribution of Enterobacteriaceae to the pathogenesis of inflammatory bowel diseases (IBDs), particularly their role in exacerbating and progressing IBD. Despite the unclear mechanism, their expansion is associated with inflammatory responses, increased levels of nitrate and oxygen, and bile acid dysmetabolism.

TLR4 signaling in the development of colitis-associated cancer and its possible interplay with microRNA-155

Jie Guo et al.

Summary: Ulcerative colitis (UC) is closely associated with an increased risk of colorectal cancer, with Toll-like receptor 4 (TLR4) playing a key role in linking oncogenic infection to colonic inflammatory and malignant processes. Overexpression of TLR4 in colitis and colitis-associated cancer (CAC) promotes tumorigenesis by facilitating cell proliferation, inhibiting apoptosis, promoting invasion and metastasis, and creating a tumor-favorable microenvironment. Studies have shown an interplay between TLR4 and miR-155 in the context of different disorders or cell lines, with a possible positive feedback loop between TLR4 and miR-155 contributing to the synergistic effect on CAC development.

Gut inflammation triggers C/EBPβ/δ-secretase-dependent gut-to-brain propagation of Aβ and Tau fibrils in Alzheimer's disease

Chun Chen et al.

Summary: Inflammation activates C/EBP beta/delta-secretase and initiates AD-associated pathologies in the gut, which then spread to the brain via the vagus nerve. Inhibition of this signaling pathway may attenuate AD-like pathologies in both the gut and the brain, restoring cognitive function. This study suggests a potential therapeutic strategy for Alzheimer's disease by targeting the gut-brain axis.

NOX1/NADPH oxidase is involved in the LPS-induced exacerbation of collagen-induced arthritis

Misaki Matsumoto et al.

Summary: The study investigated the potential role of NOX1, a non-phagocytic isoform of NADPH oxidase, in immune responses using Nox1-knockout mice. While NOX1 deficiency did not affect antibody production and asthma responses, it significantly reduced the severity and incidence of experimental arthritis. These findings suggest that LPS-inducible NOX1 in monocytes/macrophages/dendritic cells may modulate the development of arthritis exacerbated by bacterial infection.

Summary: Using PDOs, the study investigated the epithelial-specific transcriptional and secretome signatures of the ileum in CD patients, identifying disease profiles and potential druggable targets. By analyzing DE genes, candidate small molecules were found to potentially reverse different CD phenotypes, and the secretome of PDOs showed both similarity and diversity in the levels and types of soluble factors released. The study concluded that targeting the epithelium could reverse stricturing phenotype and improve outcomes.
