Validation of four cutaneous squamous cell carcinoma staging systems using nationwide data

Zoe Claire Venables et al.

Summary: This study validated the performance of four cSCC staging systems in predicting metastasis, with the BWH system demonstrating the highest specificity and discriminative ability, while the AJCC8 system showed superior negative predictive value and homogeneity. Further improvement and refinement of current cSCC staging systems are needed.

Effect of Nicotinamide in Skin Cancer and Actinic Keratoses Chemoprophylaxis, and Adverse Effects Related to Nicotinamide: A Systematic Review and Meta-Analysis

Laurence Mainville et al.

Summary: This study evaluated the effect of oral nicotinamide in preventing skin cancers through a systematic review and meta-analysis of randomized controlled trials. The results showed that nicotinamide was associated with a significant reduction in skin cancers, particularly basal cell carcinomas and cutaneous squamous cell carcinomas. However, it was also associated with an increased risk of digestive adverse effects.

Cumulative incidence and disease-specific survival of metastatic cutaneous squamous cell carcinoma: A nationwide cancer registry study

Selin Tokez et al.

Summary: This study investigated the cumulative incidence and disease-specific survival of patients with metastatic cutaneous squamous cell carcinoma (mcSCC) in the Dutch population and assessed patient-based risk factors. The results showed that approximately 2% of cSCCs metastasize, with higher risk for men, increasing age, and immunocompromised patients. The 5-year disease-specific survival for patients with mcSCC was high.

Assessment of the Diagnostic Accuracy of Baseline Clinical Examination and Ultrasonographic Imaging for the Detection of Lymph Node Metastasis in Patients With High-Risk Cutaneous Squamous Cell Carcinoma of the Head and Neck

Selin Tokez et al.

Summary: In this study, baseline ultrasonography was found to be more sensitive than clinical examination alone for detecting lymph node metastasis in patients with high-risk cutaneous SCC of the head and neck. Ultrasonography showed high sensitivity for detecting nodal metastases, especially in patients with negative baseline clinical examination results. However, the high rate of false-positive findings should be taken into consideration.

Mean lifetime survival estimates following solid organ transplantation in the US and UK

Christopher N. Graham et al.

Summary: This study explores the flexible cubic spline methodology as an alternative for estimating lifetime survival after solid organ transplant. The study found variations in mean survival among different organ transplants and highlights the importance of including long-term hazards in survival estimates. These estimates can be used by decision-makers in healthcare resource allocation decisions as a meaningful alternative to current practice.

Significant Biomarkers Identification Associated with Cutaneous Squamous Cell Carcinoma Progression

Cheng-Gang Qiu et al.

Summary: This study aimed to identify significant genes associated with cutaneous squamous cell carcinoma (CSCC) initiation and development. The results revealed that MYBL2 and TK1 were the most significant biomarkers associated with CSCC initiation and progression, and they may act as promising therapeutic targets for accurate diagnosis and treatment of CSCC.

High-Risk Cutaneous Squamous Cell Cancer of the Head and Neck: Risk Factors for Recurrence and Impact of Adjuvant Treatment

Samuel J. Trosman et al.

Summary: Despite adjuvant treatment, advanced HNcSCC has a high recurrence rate, with tumor size larger than 2 cm serving as a strong independent risk factor for recurrence. Among traditional HNcSCC risk factors, perineural invasion was most strongly associated with worse disease-free survival. The addition of chemotherapy did not show survival benefit for patients in this study.

Differential Molecular Expression Patterns Associated with Metastasis in Cutaneous Squamous Cell Carcinoma: A Systematic Review and Meta-Analysis

Patrick M. Mulvaney et al.

Summary: High expression of PD-L1, EGFR, and podoplanin in primary tumors may increase the risk of metastasis, while metastatic tissue is more likely to have high PD-L1 expression. Further studies are needed to determine whether testing for these markers can aid in prognostic estimation and predicting treatment response in cutaneous squamous cell carcinomas.

Summary: Cutaneous squamous cell carcinomas (cSCC) are among the most common non-melanoma skin cancers, with treatment depending on whether it is early stage/low risk or locally advanced/metastatic. Therapeutic approaches include surgery, radiation, and systemic therapies, with recent immunotherapeutics showing promising tumor responses in advanced cSCC.

Adjuvant therapy for high-risk cutaneous squamous cell carcinoma: A 10-year review

Jason G. Newman et al.

Summary: The standard of care for high-risk cutaneous squamous cell carcinoma involves surgical excision with clear margins, but adjuvant therapy may be considered for certain high-risk factors. However, patient selection for adjuvant therapy remains unclear due to conflicting evidence and limited research. Prognostic tools and large-cohort clinical studies are needed to determine the most appropriate therapy and patient selection for high-risk cSCC.
