Autoimmune polyendocrine syndrome type 1 (APECED) in the Indian population: case report and review of a series of 45 patients

A. Fierabracci et al.

Summary: APECED, also known as APS-1, is a rare autosomal recessive genetic disease caused by mutations in the AIRE gene. Clinical diagnosis is based on the presence of chronic mucocutaneous candidiasis, hypoparathyroidism, and primary adrenal insufficiency. Indian APECED patients may benefit from earlier diagnosis using criteria validated in North/South American patients.

The Ever-Increasing Array of Novel Inborn Errors of Immunity: an Interim Update by the IUIS Committee

Stuart G. Tangye et al.

Summary: The latest classification of inborn errors of immunity/primary immunodeficiencies by the International Union of Immunological Societies Expert Committee, published in January 2020, quickly became outdated due to the rapid pace of genetic variant discoveries in the field. To bridge the gap until the next official report in 2022, researchers have highlighted 26 additional monogenic gene defects as novel causes of immune defects.

GWAS for autoimmune Addison's disease identifies multiple risk loci and highlights AIRE in disease susceptibility

Daniel Eriksson et al.

Summary: This study conducted the first genome-wide association study on Autoimmune Addison's disease, identifying nine risk loci, including two coding variants in the Autoimmune Regulator (AIRE) gene. These findings highlight the importance of central immunological tolerance and explain a significant portion of heritability.

A distal Foxp3 enhancer enables interleukin-2 dependent thymic Treg cell lineage commitment for robust immune tolerance

Autoimmune polyendocrine syndrome type 1: an Italian survey on 158 patients

S. Garelli et al.

Summary: Autoimmune Polyglandular Syndrome type 1 (APS-1) in Italy is a rare disorder characterized by the three major manifestations and associated with various AIRE gene mutations. IFN omega Abs are markers for APS-1, while other organ-specific autoantibodies are markers for clinical, subclinical, or potential autoimmune conditions.

Genome-wide association study identifies five risk loci for pernicious anemia

Triin Laisk et al.

Summary: Pernicious anemia is a rare condition with a clear autoimmune basis, often caused by autoimmune gastritis, and associated with a higher risk of other autoimmune disorders. A genome-wide association study meta-analysis identified significant associations between pernicious anemia and genetic risk factors in genes such as PTPN22, PNPT1, HLA-DQB1, IL2RA, and AIRE.
