The Heparanase Regulatory Network in Health and Disease

Alyce J. Mayfosh et al.

Summary: The extracellular matrix plays crucial roles in maintaining tissue structure, immune defense, and acting as a reservoir for bioactive molecules, with heparan sulfate proteoglycans (HSPGs) and their component heparan sulfate being essential in these processes. Understanding the regulation of heparanase and its downstream targets is important for potential therapeutic strategies in inflammatory diseases, atherosclerosis, fibrosis, and cancer metastasis.

Rheumatoid arthritis: Extra-articular manifestations and comorbidities

Fabiana Assunta Figus et al.

Summary: Synovitis is the primary manifestation of rheumatoid arthritis (RA), but many extra-articular manifestations and comorbidities are likely due to the chronic, complex nature of the disease. Cardiovascular disease is the leading cause of death in RA patients, with increased risks of myocardial infarction and CV mortality. Other common causes of death in RA patients include respiratory disease and neurological issues, with evidence linking systemic inflammation to Alzheimer's disease and vascular dementia. In order to reduce morbidity and mortality, physicians must actively manage RA disease activity and monitor for potential risk factors and associated conditions.

Macrophages bind LDL using heparan sulfate and the perlecan protein core

Chun-yi Ng et al.

Summary: The research reveals an important role of polarized macrophage-derived proteoglycans in the pathogenesis of atherosclerosis, particularly in the retention of LDL. The study shows that proteoglycans from activated macrophages can bind LDL both through the protein core and heparan sulfate. These findings suggest that macrophages may contribute to LDL retention in atherosclerotic plaques by producing proteoglycans.

Circulating Heparan Sulfate Proteoglycans as Biomarkers in Health and Disease

Antonio Junior Lepedda et al.

Summary: Cell-surface heparan sulfate proteoglycans (HSPGs) play crucial roles in regulating cell behavior and signaling, with their soluble forms potentially serving as diagnostic and prognostic markers and targets for immunotherapy. This comprehensive review critically surveys clinical findings from the past 20 years and discusses the involvement of HSPGs in various pathologies, highlighting their potential diagnostic and therapeutic value.

Effect of Heparanase and Heparan Sulfate Chains in Hemostasis

Yona Nadir

Summary: Heparanase, the only known mammalian enzyme to degrade heparan sulfate chains, affects the hemostatic system through various mechanisms, including degrading effect, release of Syndecan-1, and enhancing tissue factor activity. In clinical settings such as cancer and pregnancy, elevated levels of Heparanase and heparan sulfate chains may increase the risk of thrombosis.

Cardiac Dysfunction in Rheumatoid Arthritis: The Role of Inflammation

Jianmin Chen et al.

Summary: Patients with rheumatoid arthritis have an increased risk of cardiovascular disease, with inflammation playing a key role. Current anti-inflammatory therapeutics used for treating RA patients may have an impact on the development of cardiovascular disease while targeting resolution of inflammation could offer an alternative strategy for controlling inflammation-driven comorbidities in RA.

Vascular cell adhesion molecule-1 in rheumatoid arthritis patients Relation to disease activity, oxidative stress, and systemic inflammation

Heba R. Salem et al.

Summary: The study revealed that RA patients had higher levels of serum VCAM-1, malondialdehyde, ESR, and C-reactive protein, while showing lower total antioxidant capacity and high-density lipoprotein levels. Serum VCAM-1 was positively correlated with disease activity, oxidative stress, and inflammatory markers, and negatively correlated with total antioxidant capacity.
