Targeting of GSDMD sensitizes HCC to anti-PD-1 by activating cGAS pathway and downregulating PD-L1 expression

Hepatocyte pyroptosis and release of inflammasome particles induce stellate cell activation and liver fibrosis

Summary: The study found that hepatocyte pyroptosis, caused by NLRP3 inflammasome activation, leads to the release of inflammasome proteins and exacerbates fibrogenesis. Inhibition of caspase-1 and gasdermin D activation can prevent pyroptosis. The released inflammasomes are taken up by hepatic stellate cells, promoting inflammation and fibrosis. These findings suggest a novel mechanism for liver injury and potential therapeutic targets for chronic liver diseases.
