Single-cell atlas of early human brain development highlights heterogeneity of human neuroepithelial cells and early radial glia

Ugomma C. Eze et al.

Summary: Through single-cell RNA sequencing, nine progenitor populations at the earliest stages of human brain development were identified, including a highly prevalent mesenchymal-like population that disappears once neurogenesis begins. Comparison with mouse progenitor populations revealed two clusters enriched in the early stages of human cortical development. Organoid systems displayed low fidelity to neuroepithelial and early radial glia cell types, but improved as neurogenesis progressed.

Summary: The study used Smart-seq2 to analyze the molecular characteristics of glomerulus-associated cells in human and mouse kidneys, revealing gene expression profiles, heterogeneity, and species differences. This helps in designing translational studies and understanding the molecular identity of renal cells.

Perspective of mesenchymal transformation in glioblastoma

Yona Kim et al.

Summary: GBM remains a challenging disease to treat due to its high intra- and inter-tumoral heterogeneity, with the mesenchymal subtype being associated with the most aggressive and treatment-resistant features. Research efforts have focused on identifying and understanding the master mesenchymal regulators and their downstream signaling pathways in order to develop more personalized therapeutic strategies.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.
