Effect of the Substituent and Amino Group Position on the Lipase-Catalyzed Resolution of γ-Amino Esters: A Molecular Docking Study Shedding Light on Candida antarctica lipase B Enantioselectivity

Summary: The kinetic resolution of N-tert-butoxycarbonyl gamma-amino methyl esters with different stereocenters was carried out using Candida antarctica lipase B in a solvent, showing differences in enantioselectivity compared to similar N-protected beta-amino esters. Molecular docking studies revealed specific regions in the enzyme cavity responsible for high selectivity in resolving carboxylic acid derivatives with beta stereocenters. The conformational diversity of gamma-amino esters led to reduced effectiveness in interactions, resulting in loss of enantioselectivity in the resolution process.
