Caveolin-3 and Arrhythmias: Insights into the Molecular Mechanisms

Miaomiao He et al.

Summary: Caveolin-3 plays a critical role in the cardiovascular system and is associated with cardiovascular diseases. Mutations in caveolin-3 can cause clinical phenotypic variations, including cardiomyopathy. These mutations affect current density and promote arrhythmia. Caveolin-3 overexpression or knockdown can contribute to the occurrence and development of arrhythmia. Additionally, caveolin-3 is related to obesity and glucose homeostasis.

Diabetes-induced changes in cardiac voltage-gated ion channels

Nihal Ozturk et al.

Summary: Diabetes affects the heart through various mechanisms, including microvascular defects, metabolic abnormalities, and electrophysiological changes, leading to diabetic cardiomyopathy (DCM) which can cause structural and functional changes in the heart. DCM progresses to cause electrical remodeling, left ventricular dysfunction, and heart failure, contributing significantly to arrhythmias and sudden cardiac death in diabetes patients.

Summary: Rhythm disturbances are life-threatening cardiovascular diseases that can be better understood through preclinical modeling of human pathophysiology, leading to effective strategies for prevention, diagnosis, and clinical management. In the vast array of rhythm disturbances, diseases of the conduction system have been extensively preclinically modeled.

Beyond pacemaking: HCN channels in sinoatrial node function

Konstantin Hennis et al.

Summary: HCN channels are essential for heart rhythm initiation and regulation, synchronizing pacemaker cells for stable heart function. Entrainment processes in the sinoatrial node play a crucial role in coordinating heart activity and regulating heart rate.

Impaired regulation of heart rate and sinoatrial node function by the parasympathetic nervous system in type 2 diabetic mice

Summary: This study found that impaired regulation of heart rate by the parasympathetic nervous system in type 2 diabetes is due to reduced responsiveness of sinoatrial node myocytes to parasympathetic agonists, which is associated with enhanced activity of RGS4.

Electrical Features of the Diabetic Myocardium. Arrhythmic and Cardiovascular Safety Considerations in Diabetes

Monica Gallego et al.

Summary: Diabetes is a chronic metabolic disease with hyperglycemia, and cardiovascular disease is a major complication leading to mortality in diabetic patients. Diabetic cardiomyopathy causes abnormal heart function and electrical remodeling, including prolonged QT interval, increasing the risk of life-threatening ventricular arrhythmias. New antidiabetic drugs must undergo thorough cardiovascular evaluation to reduce the risk of arrhythmia and sudden death in diabetic patients.
