Caspofungin suppresses zymosan-induced cytokine and chemokine release in THP-1 cells: possible involvement of the spleen tyrosine kinase pathway

Kazuhiro Itoh et al.

Summary: This study investigated the effects of the antifungal agent caspofungin (CAS) on pro-inflammatory cytokines and chemokines in zymosan-activated THP-1 cells. It was found that CAS suppressed the release of these inflammatory mediators by inhibiting the activation of Syk and its downstream signaling molecules. These results suggest that CAS may have the potential to prevent hypercytokinemia in fungal sepsis through the Syk signaling pathway.

Tumor Mutational Burden as a Predictor of Immunotherapy Response: Is More Always Better?

John H. Strickler et al.

Summary: While high tumor mutational burden (TMB-H) has shown promise as a biomarker in lung cancer, its broad applicability across all solid tumors is unclear. The FDA has approved the PD-1 inhibitor, pembrolizumab, as a therapy for all solid tumors with TMB equal to or greater than 10 mutations. However, there are risks and unanswered questions regarding the use of TMB as a biomarker.

Agonistic CD40 therapy induces tertiary lymphoid structures but impairs responses to checkpoint blockade in glioma

Luuk van Hooren et al.

Summary: This study demonstrates that the use of immunostimulatory CD40 antibodies in glioma models promotes the formation of tertiary lymphoid structures but does not improve survival and impairs the response to immune checkpoint blockade.
