Reversible Covalent Headpiece Enables Interconversion between Double- and Single-Stranded DNA-Encoded Chemical Libraries

Guixian Zhao et al.

Summary: The use of an appropriate encoding methodology is crucial when practicing DEL technology. The headpiece-based double-stranded DEL encoding method is currently the most widely used for productive DEL, but it conflicts with the versatility of single-stranded DEL applications. A novel encoding method based on a reversible covalent headpiece has been reported, allowing reversible interconversion between double- and single-stranded DNA formats, providing robust synthesis, and enabling applications in different setups.

Photoredox Deaminative Alkylation in DNA-Encoded Library Synthesis

Yurong Shen et al.

Summary: An on-DNA photoredox-mediated deaminative alkylation method has been reported for diversifying DNA-tagged acrylamide substrate with amine-derived radicals. The method allows convenient generation of radicals from sterically hindered primary amines, and can tolerate a wide range of radical precursors. It is also applicable to Boc-protected diamines, free amino acids, and aryl halides, enabling additional rounds of diversification.
