Summary: LAG-3 and PD-1 are two distinct inhibitory immune checkpoints that contribute to T-cell exhaustion. The combination of relatlimab and nivolumab has been shown to be safe and effective in previously treated melanoma patients, and this study aimed to evaluate its safety and activity in previously untreated melanoma patients. The results demonstrated that the combination therapy provided a greater benefit in terms of progression-free survival compared to nivolumab alone, without any new safety signals.

We also show that eEF2K plays a vital role in regulating the tumor immune microenvironment by controlling PD-L1 expression. In addition, combining eEF2K inhibition with ICB therapy may provide a potential combination therapeutic strategy.

Summary: This study investigated the regulation by immune checkpoint in preventing tissue damage caused by ischemia reperfusion (I/R) and the effects of pentoxifylline (PTX) as a protective drug on gene and protein expressions. Results showed significant differences in the expression levels of SNHG14, miR-1206, and PAG1 in I/R and PTX treated groups. PTX doses effectively suppressed CTLA4 development and reduced the number of regulatory T cells. SNHG14 and miR-1206 showed high diagnostic sensitivity and specificity in I/R.
