Metabolic modulation of immune checkpoints and novel therapeutic strategies in cancer

Yi Wang et al.

Summary: Cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) or programmed death-1 (PD-1)/programmed death-ligand 1 (PD-L1)-based immune checkpoint inhibitors (ICIs) have shown significant improvements in overall survival of certain cancer patients, but resistance to immunotherapy remains a challenge. This article examines the metabolic modulation of tumor immunity and immune checkpoints in the tumor microenvironment (TME) and discusses potential metabolism-based therapeutic strategies. Understanding the link between tumor metabolism and immunotherapy is crucial for cancer treatment and research.

CMTM6 and PD-1/PD-L1 overexpression is associated with the clinical characteristics of malignancy in oral squamous cell carcinoma

Songtao Zhang et al.

Summary: The study showed that the expression of CMTM6 and PD-1/PD-L1 was significantly higher in OSCC tissues compared to paracancerous tissues. PD-L1 expression was associated with clinical stage and lymph node metastasis in OSCC. qRT-PCR results confirmed the relationship between mRNA and protein expression.

Summary: This study provides evidence for the prognostic significance of CD8+ and CD4+ TILs in HNSCC, with variations in tumor subsite needing further investigation. TILs may serve as predictive biomarkers for risk stratifying patients and further research is needed in the application of immune-checkpoint inhibitors.

High Expression of Hyaluronan-Mediated Motility Receptor Predicts Adverse Outcomes: A Potential Therapeutic Target for Head and Neck Squamous Cell Carcinoma

Tianzhu Lu et al.

Summary: The study found that HMMR is highly expressed in HNSCC tumor tissue, and high HMMR mRNA expression is an independent prognostic factor in TCGA and GSE41613 data. The methylation level of HMMR is negatively correlated with its expression, with patients having low methylation levels showing worse overall survival. HMMR may be associated with biological functions related to the KARS, EMT, and G2M checkpoint pathways, interferon-gamma, and interferon-alpha responses.

DCBLD1 is associated with the integrin signaling pathway and has prognostic value in non-small cell lung and invasive breast carcinoma

Guillaume B. Cardin et al.

Summary: Germline single nucleotide polymorphisms in the promoter region of the DCBLD1 gene are associated with non-smoking cases of both non-small cell lung carcinoma (NSCLC) and human papillomavirus-negative head and neck cancer, with high DCBLD1 expression linked to worse overall survival. Patients with high DCBLD1 expression show an upregulation of the integrin signaling pathway.

The Role of Oncogenes and Redox Signaling in the Regulation of PD-L1 in Cancer

Christophe Glorieux et al.

Summary: PD-L1 is a crucial immune checkpoint molecule in cancer that is regulated by genetic, epigenetic, and transcriptional factors as well as posttranscriptional and posttranslational processes. Understanding these regulatory mechanisms is important for developing new therapeutic strategies to overcome tumor immune evasion.

High SEC61G expression predicts poor prognosis in patients with Head and Neck Squamous Cell Carcinomas

Leifeng Liang et al.

Summary: The study revealed that SEC61G is significantly overexpressed in HNSCC tissues, and its high expression is significantly correlated with various clinicopathologic features. ROC curves demonstrated the significant diagnostic ability of SEC61G for HNSCC. Kaplan-Meier survival analysis showed that HNSCC patients with high SEC61G expression had a poorer prognosis. Multivariate analysis indicated that SEC61G is independently associated with overall survival. Function enrichment analysis suggested that SEC61G is involved in pathways related to translation and regulation of SLITs/ROBOs expression, which may impact HNSCC progression and prognosis.
