Increased Sensitivity of SARS-CoV-2 to Type III Interferon in Human Intestinal Epithelial Cells

Camila Metz-Zumaran et al.

Summary: This study demonstrates that both type I and III interferons can inhibit SARS-CoV-2 infection in human intestinal cells, but type III interferons exhibit stronger antiviral activity. Shorter treatment times and lower concentrations are required for type III interferons to effectively reduce virus load. Furthermore, type III interferons can significantly inhibit SARS-CoV-2 even 4 hours post-infection and induce a long-lasting antiviral effect in intestinal epithelial cells.

SARS-CoV-2 pathogenesis

Mart M. Lamers et al.

Summary: This Review explores the recent clinical and experimental advancements in understanding the pathogenesis of SARS-CoV-2, its interactions with host cells, and the role of the immune system in the development of severe disease, with a focus on the mechanisms underlying COVID-19-associated ARDS. The findings suggest that SARS-CoV-2 can infect the lower respiratory tract, causing alveolar damage and dysfunctional immune responses, leading to inflammation and immunopathology. These insights may contribute to the development of new therapeutic interventions against COVID-19.

Targeted Profiling of Immunological Genes during Norovirus Replication in Human Intestinal Enteroids

Jenny C. M. Chan et al.

Summary: Norovirus is the major cause of acute gastroenteritis worldwide but its pathogenesis and immune response are still unclear due to the lack of a robust virus culture system. In this study, productive norovirus replications were achieved in 75% of inoculations using secretor-positive Chinese human intestinal enteroid (HIE) lines. The upregulation of immune-related genes CXCL10 and IFI44L was positively correlated with norovirus RNA replication levels, suggesting HIE as a potential model for future virus-host interaction studies.

Controlling the polarity of human gastrointestinal organoids to investigate epithelial biology and infectious diseases

Julia Y. Co et al.

Summary: The polarity of gastrointestinal organoids is reversed to study epithelial biology and host-microbe interactions, allowing increased access to the apical surface while preserving functions. The method quickly establishes an outward-facing apical surface, maintaining epithelial integrity, secretory and absorptive functions, and enabling differentiation control. This technique enhances the possibilities of organoid use in gastrointestinal and respiratory health and disease research by controlling organoid polarity.
