Interconnections between Inflammageing and Immunosenescence during Ageing

Thibault Teissier et al.

Summary: Acute inflammation is necessary for healing, but chronic inflammation is harmful. Inflammageing refers to age-associated chronic inflammation, which is not solely caused by chronological age, but also by biological ageing and increased mortality risk. Cellular senescence and immunosenescence are also involved in inflammageing. This review explores the sources and consequences of inflammageing, focusing on the contribution of cellular senescence and alterations in the ageing immune system. It also discusses potential interventions to reduce inflammageing and improve later life health.

DNA Methylation and Immune Cell Markers Demonstrate Evidence of Accelerated Aging in Patients with Chronic Hepatitis B Virus or Hepatitis C Virus, with or without Human Immunodeficienct Virus Co-infection

Yevgeniy Gindin et al.

Summary: Patients with chronic viral hepatitis have accelerated epigenetic aging, with immune markers defining biological age and potential to assess therapeutic intervention effects on age acceleration. Age acceleration in patients with chronic hepatitis B was associated with higher epigenetic age, while in patients with chronic hepatitis C, it was linked to liver fibrosis or HIV co-infection. The use of immune markers to define age acceleration was correlated with DNA methylation results.

Immunosenescence Study of T Cells: A Systematic Review

Ivon Johanna Rodriguez et al.

Summary: Aging leads to alterations in immune response, termed immunosenescence, characterized by decreased naive T cells and increased expression of CD57 and KLRG1 in memory T cells in older adults. Proinflammatory cytokines increase and telomere length decrease in old adult T cells. The heterogeneity and biases among studies hinder robust statistical analysis for identifying flow cytometry markers of T cell immunosenescence phenotype.

Targeting and clearance of senescent foamy macrophages and senescent endothelial cells by antibody-functionalized mesoporous silica nanoparticles for alleviating aorta atherosclerosis

Le Minh Pham et al.

Summary: This study found that CD9 antibody-modified nanoparticles showed high cellular uptake in vitro models, reducing oxidative stress and inflammation in senescent cells, and improving cell viability. In vivo experiments demonstrated that these nanoparticles effectively targeted senescent areas, delivering anti-senescence drugs to atherosclerotic plaques and alleviating the progression of atherosclerosis. This suggests a novel and precise therapeutic approach for treating atherosclerosis by targeting senescent cells.

Murilo Delgobo et al.

Summary: The cardiovascular and immune systems undergo significant changes with aging, and an accumulation of CD4(+) T cells in elderly individuals may contribute to myocardial aging and heart disease progression. By studying young mice with expanded CD4(+) T cell compartments, researchers found that these cells can differentiate and impact the heart, suggesting a link between immunological senescence and age-related cardiac deterioration.

Summary: Recent evidence suggests that fibrotic liver injury in chronic hepatitis C patients is related to cellular senescence in damaged liver tissue, but it is still unknown how senescence impacts HCV replication. A study found that a cyclin-dependent kinases inhibitor induced cellular senescence in hepatoma cells and accelerated viral replication. Additionally, the antiviral effects of host-targeting agents varied when senescence was induced in cells.

Age and CMV-Infection Jointly Affect the EBV-Specific CD8+ T-Cell Repertoire

Josien Lanfermeijer et al.

Summary: CD8(+) T cells play a crucial role in guarding against viral infections, but changes in the T-cell pool with age can lead to diminished responses. Both a decrease in T-cell numbers and T-cell receptor diversity are thought to contribute to this age-related decline in immune function. Latent CMV infection and aging may impact the diversity of T-cell repertoires, affecting responses to other antigens.
