Effects of aneuploidy on cell behaviour and function

Rong Li et al.

Summary: Aneuploidy, characterized by deviations in the copy number of chromosomes, affects organisms from early development through to aging and is a main cause of human pregnancy loss and cancer. Recent studies have increased our understanding of its mechanisms and how it can be both beneficial and detrimental, depending on the karyotype and external cues. These insights shed light on its roles in human pathogenesis and genome evolution.

Genome Chaos, Information Creation, and Cancer Emergence: Searching for New Frameworks on the 50th Anniversary of the War on Cancer

Julie Heng et al.

Summary: Despite significant progress in understanding the molecular mechanisms of cancer in the past 50 years, clinical benefits are still lacking. Therefore, it is important to pause and reflect on the biological meaning of the existing data and integrate new ideas in cancer biology to advance research.

Location-specific cell identity rather than exposure to GI microbiota defines many innate immune signalling cascades in the gut epithelium

Ozge Kayisoglu et al.

Summary: The study demonstrates extensive spatial organization of innate immune signaling components along the cephalocaudal axis in the GI tract. This organization is largely determined before birth and independent of exposure to commensal gut microbiota. Gastric epithelial organoids can sense LPS from both the basal and apical sides.

Massively Parallel Sequencing of Esophageal Brushings Enables an Aneuploidy-Based Classification of Patients With Barrett's Esophagus

Christopher Douville et al.

Summary: RealSeqS analysis of esophageal brushings provides a practical and sensitive method to determine aneuploidy in patients with Barrett's esophagus. It identifies specific chromosome changes that occur early in the disease and others that occur late and mark disease progression. The clinical implications of this approach can now be tested in prospective trials.

The unfolded protein response links tumor aneuploidy to local immune dysregulation

Su Xian et al.

Summary: Aneuploidy is associated with poor prognosis in cancer types, and the unfolded protein response (UPR) is proposed as a mechanistic link between aneuploidy and immune dysregulation in the tumor microenvironment, showing increased activity of multiple UPR branches in response to aneuploidy.

Tumor morphology and location associate with immune cell composition in pleomorphic sarcoma

Rosanna L. Wustrack et al.

Summary: This study provides an immune-based analysis of the tumor microenvironment in UPS patients, showing different composition of tumor infiltrating lymphocytes based on size and tumor depth. In particular, an inverse correlation between tumor-infiltrating CD8 + T cells and UPS tumor size, as well as a positive correlation between tumor-infiltrating CD8 + T cells and overall survival, were identified. Additionally, an association between anatomical location (deep or superficial) and frequency of CD4 + PD1(hi) infiltrating T cells in UPS tumors was demonstrated.

Genetic and non-genetic clonal diversity in cancer evolution

James R. M. Black et al.

Summary: This article discusses the importance of functional and non-functional intra-tumour heterogeneity (ITH) in cancer evolution. It emphasizes the need to consider genetic and non-genetic factors in tumor evolution, and highlights the role of systems-biology analytical approaches in understanding the functional ITH. Ultimately, deepening our understanding of tumour evolution through functional ITH will lead to the development of novel therapies and improved outcomes for patients.

Clonal expansion in non-cancer tissues

Nobuyuki Kakiuchi et al.

Summary: Clonal expansion in phenotypically normal or non-cancer tissues is commonly associated with aging, environmental insults, and chronic inflammation, but it does not necessarily indicate cancer development. This review discusses recent findings on clonal expansion in these tissues and their biological significance in cancer development, aging, and inflammatory diseases.

Aneuploidy as a promoter and suppressor of malignant growth

Anand Vasudevan et al.

Summary: Aneuploidy, although frequent in human tumors, is not always a driver of cancer development and can have tumor-suppressive effects. The specific consequences of aneuploidy are context-dependent and influenced by genetic and environmental factors.
