CD8+ T cells and fatty acids orchestrate tumor ferroptosis and immunity via ACSL4

Peng Liao et al.

Summary: T cell-derived interferon gamma (IFN γ) in combination with arachidonic acid (AA) induces immunogenic tumor ferroptosis, serving as a mechanism for CD8(+) T cell-mediated tumor killing. This process involves IFN γ stimulation of ACSL4 and alteration of tumor cell lipid pattern.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Selective Targeting of Cancer Cells by Copper Ionophores: An Overview

Valentina Oliveri

Summary: Conventional cancer therapies face challenges due to off-target effects, leading to the need for new therapeutic agents with increased selectivity, reduced side effects, and the ability to overcome cancer cell resistance targeting cancer stem cells. Some copper ionophores show promise in inducing cuproptosis in cancer cells with greater selectivity.

Mitochondrial copper depletion suppresses triple-negative breast cancer in mice

Liyang Cui et al.

Summary: The research developed a mitochondria-targeted copper-depleting nanoparticle that effectively inhibited the growth of triple-negative breast cancer cells and improved survival rates in mouse models. By depriving copper within cancer cells, this approach induced metabolic changes and apoptosis, highlighting its potential clinical relevance.

Sorafenib induces mitochondrial dysfunction and exhibits synergistic effect with cysteine depletion by promoting HCC cells ferroptosis

Yanchun Li et al.

Summary: The study reveals that sorafenib inhibits the viability, proliferation, and migration of HCC cells by inducing ferroptosis, which can be reversed by glutathione supplementation to reduce mitochondrial reactive oxygen species and lipid peroxides accumulation, enhancing HCC cells' vulnerability to sorafenib.

Acquired deficiency of peroxisomal dicarboxylic acid catabolism is a metabolic vulnerability in hepatoblastoma

Huabo Wang et al.

Summary: Metabolic reprogramming in transformed cells provides them with proliferative and survival advantages, but therapeutic applications have been limited by small differences in metabolic pathways and cancer adaptation. However, experiments have shown that downregulation of Ehhadh in hepatoblastoma makes them more susceptible to dietary intervention with C12, leading to longer survival and potential resistance development. This research suggests the feasibility of non-toxic dietary supplementation with C12 or DDDA, which may be compatible with standard chemotherapies.

CD36-mediated ferroptosis dampens intratumoral CD8+ T cell effector function and impairs their antitumor ability

Xingzhe Ma et al.

Summary: Understanding the mechanisms of how T cells become dysfunctional in a tumor microenvironment is crucial for cancer immunotherapy. This study found that CD36 expression in tumor-infiltrating CD8(+) T cells, induced by TME cholesterol, is associated with tumor progression and poor survival, and that genetic ablation of Cd36 in these T cells leads to enhanced tumor eradication. Targeting CD36 or inhibiting ferroptosis could restore T cell function and enhance antitumor efficacy, especially in combination with anti-PD-1 antibodies.

Cirrhotic Liver of Liver Transplant Recipients Accumulate Silver and Co-Accumulate Copper

Jaroslaw Poznanski et al.

Summary: Silver-based materials are extensively used in clinical medicine, with silver nanoparticles being released into the bloodstream through silver-containing dressings. A study found significant silver accumulation in both healthy and diseased liver biopsies, particularly in patients with hepatic pathologies. The presence of silver in cirrhotic livers, which is correlated with copper levels, may be a result of the widespread application of silver in clinical settings and could have important implications for public health due to the potential toxicity of silver nanoparticles.

Murine deficiency of peroxisomal l-bifunctional protein (EHHADH) causes medium-chain 3-hydroxydicarboxylic aciduria and perturbs hepatic cholesterol homeostasis

Pablo Ranea-Robles et al.

Summary: Peroxisomes play a crucial role in the beta-oxidation of dicarboxylic acids, with deficiency of the bifunctional protein EHHADH leading to medium-chain 3-hydroxydicarboxylic aciduria. The products of DCA metabolism can be transported to mitochondria for further metabolism through stable isotope tracing methodology.

Role of Copper on Mitochondrial Function and Metabolism

Lina M. Ruiz et al.

Summary: Copper is crucial for various life processes such as energy metabolism and oxidative stress management in eukaryotic organisms. Dysregulation of copper levels can lead to diseases like anemia and cancer, prompting research into potential therapeutic interventions targeting copper metabolism. This review explores the role of copper in cellular function, metabolic reprogramming, and cancer biology, highlighting its importance in the field of regenerative medicine and oncology.
