Single-cell RNA sequencing reveals the cellular heterogeneity of aneurysmal infrarenal abdominal aorta

Guizhen Zhao et al.

Summary: This study characterized the heterogeneity and cellular responses of vascular smooth muscle cells (VSMCs) and monocytes/macrophages during abdominal aortic aneurysm (AAA) progression, providing insights into their function and the regulation of AAA onset and progression. The results identified AAA disease-relevant transcriptional signatures of vascular cells in the infrarenal abdominal aortas (IAAs) from C57BL/6J mice, revealing multiple cell types contributing to AAA.

Exosomal miR-21 secreted by IL-1β-primed-mesenchymal stem cells induces macrophage M2 polarization and ameliorates sepsis

Mengying Yao et al.

Summary: This study characterized the effects of miR-21 delivered by exosomes from MSCs on macrophage polarization in a mouse sepsis model. Beta MSC-derived exosomes induced M2-like polarization of macrophages more effectively, improving survival rates in septic mice. Exosomal miR-21 was identified as a key mediator of the immunomodulatory properties of MSCs induced by IL-1 beta pretreatment, providing a potential therapeutic basis for sepsis.

Plasminogen activator inhibitor-1 reduces cardiac fibrosis and promotes M2 macrophage polarization in inflammatory cardiomyopathy

Christian Baumeier et al.

Summary: The study showed that elevated levels of PAI-1 in high-grade DCMi patients are associated with reduced cardiac fibrosis, decreased TGF-beta levels, and fewer myofibroblasts. Additionally, high-grade DCMi patients exhibited a higher proportion of M2 macrophages compared to M1 macrophages, suggesting a role of PAI-1 in macrophage polarization in inflammatory cardiomyopathies.

Mer regulates microglial/macrophage M1/M2 polarization and alleviates neuroinflammation following traumatic brain injury

Haijian Wu et al.

Summary: Mer plays an important role in regulating microglial/macrophage M1/M2 polarization and neuroinflammation following TBI, potentially serving as a therapeutic target for intervention. Activation of the STAT1/SOCS1/3 pathway by Mer contributes to its regulatory function in this process.

High-fat diet-induced upregulation of exosomal phosphatidylcholine contributes to insulin resistance

Anil Kumar et al.

Summary: This study reveals that lean mice become insulin resistant after being administered exosomes isolated from the feces of obese mice fed a high-fat diet or from patients with type II diabetes. The altered lipid composition of exosomes from high-fat diet-fed mice affects the uptake by macrophages and hepatocytes, leading to inhibition of the insulin signaling pathway. Exosome-derived phosphatidylcholine activates AhR, inhibiting the expression of genes essential for activation of the insulin signaling pathway.

Functional delivery of lncRNA TUG1 by endothelial progenitor cells derived extracellular vesicles confers anti-inflammatory macrophage polarization in sepsis via impairing miR-9-5p-targeted SIRT1 inhibition

Wentao Ma et al.

Summary: This study reveals that EPCs derived EVs transmit TUG1 to alleviate sepsis by promoting macrophage M2 polarization. Additionally, TUG1 can competitively bind to miR-9-5p to upregulate SIRT1 expression, leading to the inhibition of miR-9-5p-mediated SIRT1 inhibition, which contributes to a more comprehensive understanding of the pathogenesis of sepsis.

Myocardial ischemia-reperfusion induced cardiac extracellular vesicles harbour proinflammatory features and aggravate heart injury

Xinyu Ge et al.

Summary: The study revealed that EVs released by the heart during ischemia-reperfusion exacerbate heart injury, while inhibiting EVs with GW4869 can reduce the damage. It was further demonstrated that IR-EVs promote M1-like polarization of macrophages and increase the expression of inflammatory cytokines.

Summary: Kidney macrophages play crucial roles in the pathogenesis of kidney diseases, with diverse functions ranging from clearing pathogens to promoting fibrosis. Their origins and proliferation are key factors in the accumulation of macrophages after kidney injury. Understanding the mechanisms driving macrophage heterogeneity and function changes could help in the development of targeted therapies for kidney diseases.

Adipose Extracellular Vesicles: Messengers From and to Macrophages in Regulating Immunometabolic Homeostasis or Disorders

Summary: Macrophages play critical roles in regulating immunometabolic homeostasis in adipose tissue by cooperating with adipocytes, ADSCs, and other cells through extracellular vesicles (EVs). Adipose EVs actively participate in the interactions between macrophages, adipocytes, and ADSCs under healthy or obese conditions, influencing immunometabolism significantly.

Summary: The study demonstrated that exosomal miR-30d-5p from PMNs contributed to sepsis-related ALI by inducing M1 macrophage polarization and priming macrophage pyroptosis through activating NF-kappa B signaling. These findings suggest a novel mechanism of PMN-M phi interaction in sepsis-related ALI, which may provide new therapeutic strategies in sepsis patients.
