Whole-genome sequencing of East Asian lung cancers reveals new germline pathogenic variants

Semanti Mukherjee et al.

Summary: In this study, Wang et al. used whole-genome sequencing to comprehensively analyze common and rare variants associated with lung cancer risk in individuals with East Asian ancestry. Their findings suggest that rare promoter variants in the BRCA2 gene are associated with an increased risk of lung cancer.

Thyroid Hormone and Deiodination in Innate Immune Cells

Anne H. van der Spek et al.

Summary: Thyroid hormone has been identified as a key factor in determining innate immune cell function, with recent evidence showing a causal relationship between cellular thyroid hormone status and innate immune cell function. This suggests that thyroid hormone metabolism may partially determine the clinical susceptibility to infections.

Global characterization of macrophage polarization mechanisms and identification of M2-type polarization inhibitors

Lizhi He et al.

Summary: Macrophages undergoing M1 versus M2-type polarization exhibit significant differences in cell metabolism, cellular functions, and signaling pathways, especially in metabolic pathways. Drug screens have identified inhibitors that selectively block M2-type polarization, with MEK signaling being found necessary for M2-type polarization in a proof-of-principle study using comprehensive proteomics and phosphoproteomics datasets.

Summary: This study describes the significant role of thyroid hormones and their nuclear receptors in intestinal development and homeostasis, and their controversial involvement in intestinal carcinogenesis. It also presents new data on their functional interaction with the WNT signaling pathway, providing new insights for translational research.

The J2-Immortalized Murine Macrophage Cell Line Displays Phenotypical and Metabolic Features of Primary BMDMs in Their M1 and M2 Polarization State

Iolanda Spera et al.

Summary: The study validated an in-house generated immortalized macrophage cell line from BMDMs by comparing its functional and metabolic characteristics to primary macrophages, showing similar features. This immortalized cell line can be used as a suitable model for evaluating how perturbations can influence the phenotypical and functional features of murine macrophages in vitro.

Thyroid Hormone Receptor Beta as Tumor Suppressor: Untapped Potential in Treatment and Diagnostics in Solid Tumors

Cole D. Davidson et al.

Summary: Dysregulation of the thyroid hormone receptor beta (TR beta) is a common characteristic in many solid and endocrine-related tumors, and despite being recognized as a tumor suppressor, the exact mechanisms by which TR beta regulates tumor growth remain unclear. Studies show that TR beta activation can induce apoptosis, reduce an aggressive phenotype, and slow tumor growth through modulation of various transcriptional networks. Additionally, the presence of TR beta is associated with positive prognosis and response to therapeutics in certain types of cancers, highlighting its potential as a diagnostic and therapeutic target.
