Reprogramming NK cells and macrophages via combined antibody and cytokine therapy primes tumors for elimination by checkpoint blockade

Chensu Wang et al.

Summary: The AIP treatment demonstrated in this study activates NK cells and macrophages, leading to transcriptional reprogramming, rapid cancer cell killing, and normalization of tumor vasculature, facilitating further immune infiltration. This innate cell-activating therapy initiates critical steps towards a self-sustaining cycle of T cell priming driven by ICB.

Cancer therapy in mice using a pure population of CD8+ T cell specific to the AH1 tumor rejection antigen

Marco Stringhini et al.

Summary: The study suggests that T cells with single antigen specificity may not be sufficient for effective tumor therapy. In an experiment using autologous T cells targeting a retroviral peptide, the T cells were able to kill tumor cells and inhibit tumor growth in vitro, but failed to induce complete tumor regression in mice, possibly due to the limited survival of injected T cells in vivo. This indicates that future therapeutic strategies using autologous T cells may need to enhance tumor-homing and survival properties of cancer-specific T cells.

Immunotherapy-induced antibodies to endogenous retroviral envelope glycoprotein confer tumor protection in mice

Byong H. Kang et al.

Summary: After curative immunotherapy of B16F10 tumors, around 60% of mice develop a strong antibody response against cell-surface tumor antigens, providing prophylactic protection against tumor formation. The majority of this humoral response is directed against the envelope glycoprotein of a murine endogenous retrovirus. Prophylactic vaccination against this protein can protect naive mice from tumor establishment following subcutaneous inoculation with B16F10 cells.

Human Endogenous Retrovirus Reactivation: Implications for Cancer Immunotherapy

Annacarmen Petrizzo et al.

Summary: Human endogenous retroviruses (HERVs) are integrated in our genome and play a crucial role in cancer immunotherapy. Reactivation of HERVs in cancer cells can lead to antiviral defense response, production of immune targets, and implications for therapeutic targets or prognosis.

Endogenous retroviruses in the origins and treatment of cancer

Natasha Jansz et al.

Summary: Endogenous retroviruses (ERVs) are emerging as promising therapeutic targets in cancer, coordinating gene expression networks and promoting interferon responses. Despite being associated with cancer, autoimmune, and neurodegenerative diseases, harnessing ERV-mediated inflammation may sensitize tumors to immunotherapy.
