A positive feed-forward loop between Fusobacterium nucleatum and ethanol metabolism reprogramming drives laryngeal cancer progression and metastasis

Chi-Yao Hsueh et al.

Summary: Alcohol consumption affects the laryngeal microbiota and high enrichment of Fusobacterium nucleatum in laryngeal squamous cell cancer (LSCC) is associated with poor prognosis. Airway Fusobacterium is Associated with Poor Response to Immunotherapy in Lung Cancer

Shujuan Chu et al.

Summary: This study explores the potential impact of the lung microbiome on the sustained clinical responses to immunotherapy in lung cancer patients. The results show that bacterial load and microbial diversity were reduced after anti-PD-1 treatment. In addition, the levels of Fusobacterium were decreased in responders post treatment, and patients with higher abundance of Fusobacterium prior to treatment were more likely to have a poor response to anti-PD-1 therapy.

Fusobacterium nucleatum Accelerates Atherosclerosis via Macrophage-Driven Aberrant Proinflammatory Response and Lipid Metabolism

Jieyu Zhou et al.

Summary: This study reveals the pathogenic mechanisms of atherosclerosis induced by Fusobacterium nucleatum and provides new insights into the prevention and treatment of atherosclerosis. F. nucleatum invaded aortic tissues and increased the progression of atherosclerotic lesions. It also changed plaque composition into a less stable phenotype and facilitated cholesterol phagocytosis and accumulation in macrophages.

Fusobacterium nucleatum is associated with inflammation and poor survival in early-stage HPV-negative tongue cancer

Sanket Desai et al.

Summary: This study provides a comprehensive analysis of infectious pathogens in various types of tumors, revealing known cancer-associated pathogens as well as a significant prevalence of Fusobacterium in head and neck tumors. The Fusobacterium-high subgroup of head and neck tumors is mutually exclusive to human papillomavirus, and exhibits characteristics such as overexpression of inflammation-related miRNAs, elevated innate immune cell fraction, and nodal metastases. A higher burden of Fusobacterium is associated with poor survival, nodal metastases, and extracapsular spread in tongue tumors, defining a distinct subgroup of head and neck cancer.

Fusobacterium nucleatum confers chemoresistance by modulating autophagy in oesophageal squamous cell carcinoma

Yang Liu et al.

Summary: F. nucleatum infection was found to reduce chemotherapeutic response in ESCC patients, by modulating autophagy to induce resistance against 5-FU, CDDP, and Docetaxel. Targeting F. nucleatum during chemotherapy may lead to improved therapeutic outcomes for ESCC patients.

A JAK/STAT-mediated inflammatory signaling cascade drives oncogenesis in AF10-rearranged AML

Bo-Rui Chen et al.

Summary: Leukemias with AF10/MLLT10 gene fusions have a poor prognosis and currently lack targeted therapies. Research demonstrates that AF10 fusions activate JAK/STAT-mediated inflammatory signaling through recruitment of JAK1 kinase, and inhibition of this signaling pathway shows promising anti-cancer effects in AF10 fusion AML models. This study identifies JAK1 as a potential therapeutic target in AF10-rearranged leukemias.

Local and systemic mechanisms linking periodontal disease and inflammatory comorbidities

George Hajishengallis et al.

Summary: Periodontitis is causally linked to the development of other chronic inflammatory diseases outside the oral mucosa, and intervention studies have shown that local treatment of periodontitis can ameliorate surrogate markers of comorbid conditions. Experimental animal studies have further strengthened the potential causal link by establishing biologically plausible and clinically consistent mechanisms. Understanding the impact of disseminated periodontal pathogens and systemic inflammation associated with periodontitis on extra-oral pathologies may lead to new therapeutic options to reduce the risk of periodontitis-associated comorbidities.
