Exploring the NK cell platform for cancer immunotherapy

Jacob A. Myers et al.

Summary: NK cells have innate potential to kill cancer cells, leading to the development of various NK cell-based therapies for cancer treatment. The main focus of NK cell therapy research is on optimizing sources of therapeutic NK cells and enhancing their cytotoxicity and persistence in the body.

Pan-cancer single cell landscape of tumor-infiltrating T cells

Liangtao Zheng et al.

Summary: This study constructed a pan-cancer atlas of T cells across 21 cancer types, revealing different composition patterns and state-transition paths of T cells. The correlation between certain T cell populations and patient characteristics sheds light on possible determinants of the tumor microenvironment, providing new insights into T cell immunity and precision immunotherapy.

Immunotherapy for sarcomas: new frontiers and unveiled opportunities

Harsimrat Kaur Birdi et al.

Summary: Sarcomas, a rare malignancy of mesenchymal tissues with over 60 subtypes, pose challenges in treatment due to disease heterogeneity. While traditional therapies are effective in early stages, immunotherapy is showing promise for advanced sarcoma treatment, sparking interest in further research and application.

Landscape of innate lymphoid cells in human head and neck cancer reveals divergent NK cell states in the tumor microenvironment

Uriel Y. Moreno-Nieves et al.

Summary: Despite the reported antitumor functions of NK cells, their contribution to tumor control in humans remains controversial due to incomplete understanding of the NK cell states within the tumor microenvironment (TME). This study demonstrates that peripheral circulating NK cells differentiate into two divergent pathways within the TME, resulting in different end states, with one having potent in vivo antitumor activity. This finding provides insight into the origin of certain NK cell states within the TME and identifies specific NK cell phenotypes with the greatest antitumor activity.

A TLR4 agonist improves immune checkpoint blockade treatment by increasing the ratio of effector to regulatory cells within the tumor microenvironment

A. Farias et al.

Summary: BLS activates dendritic cells to induce secretion of pro-inflammatory cytokines and chemokines, but its therapeutic effect diminishes in late stages of tumor growth; administration of BLS increases serum IFN-gamma levels and immune cell infiltration in the tumor, and combined with PD-1 blockade therapy, it enhances the outcome of monotherapies.

Comparative Immunogenomics of Canine Natural Killer Cells as Immunotherapy Target

Alicia A. Gingrich et al.

Summary: Dogs serve as an important model for studying NK cell biology and immunotherapy, with their gene expression profiles providing insight into NK cell functions across different conditions and species. Comparative analysis reveals that canine NK cells are more similar to human NK cells at the gene level, highlighting the translational relevance of these findings. This study helps bridge the gap in NK research by defining canine NK cell gene expression and comparing it to other species.
