Subtherapeutic Photodynamic Treatment Facilitates Tumor Nanomedicine Delivery and Overcomes Desmoplasia

Marta Overchuk et al.

Summary: The study demonstrates that subtherapeutic photodynamic priming (PDP) enhances nanoparticle accumulation in tumors, reduces tumor collagen content, and decreases extracellular matrix density. Combining PDP with Doxil effectively delays tumor growth and improves therapeutic outcomes.

Pharmacologic Normalization of Pancreatic Cancer-Associated Fibroblast Secretome Impairs Prometastatic Cross-Talk With Macrophages

Remi Samain et al.

Summary: The study suggests that SOM230 has the potential to be an effective anti-metastatic therapy in PDA by regulating stromal activities, reducing inflammatory responses, and improving the immunosuppressive tumor microenvironment. This pharmacotherapy shows promise in reshaping the tumor environment and enhancing treatment outcomes for pancreatic cancer patients.

EUS-guided verteporfin photodynamic therapy for pancreatic cancer

Yuri Hanada et al.

Summary: This study evaluated the feasibility of EUS-guided verteporfin PDT in ablating nonresectable locally advanced pancreatic cancer (LAPC). Results showed promise for this minimally invasive ablative therapy in select patients, with visible tumor necrosis within 48 hours post-treatment. Patient enrollment and data collection are ongoing.

What NIR photodynamic activation offers molecular targeted nanomedicines: Perspectives into the conundrum of tumor specificity and selectivity

Chanda Bhandari et al.

Summary: Near infrared (NIR) photodynamic activation plays a crucial role in cutting-edge anti-cancer nanomedicines, with molecular targeted photonanomedicines (mt-PNMs) specifically targeting tumor tissue treatment with high precision. However, mt-PNMs are complex and face challenges, while providing opportunities to enhance ligand targeting effects. Both tumor specificity and selectivity are discussed with a focus on NIR photoactivable biomimetic nanotechnologies for safer and more effective photodynamic anti-tumor regimens.

CT radiomic features of photodynamic priming in clinical pancreatic adenocarcinoma treatment

Phuong Vincent et al.

Summary: This study investigated the potential of radiomic analysis using CT scans to identify PDT-induced necrosis areas and quantify PDP effects in pancreatic tumors. The results showed significant differences in intensity and texture-based features in areas with PDT-induced necrosis and in surrounding tissues affected by PDP. Texture features extracted from CT scans could be utilized for clinical diagnostic prediction and assessment of PDT and PDP effects in pancreatic tumors.

Photodynamic priming with triple-receptor targeted nanoconjugates that trigger T cell-mediated immune responses in a 3D in vitro heterocellular model of pancreatic cancer

Pushpamali De Silva et al.

Summary: Studies have shown that photodynamic priming (PDP) can transiently change the tumor microenvironment and enhance immune stimulation by increasing tumor permeability and inducing immunogenic cell death. Using targeted photoimmuno-nanoconjugate (TR-PINs)-mediated PDP, researchers were able to demonstrate increased cytotoxicity in pancreatic ductal adenocarcinoma (PDAC) cells and enhanced T cell activation, leading to significant tumor cell death and improved immunogenicity.

Membrane composition is a functional determinant of NIR-activable liposomes in orthotopic head and neck cancer

Mina Guirguis et al.

Summary: This study evaluates the impact of different chemical properties and conformations on the functionality of NIR-activable liposomes, showing that membrane composition is crucial for their effectiveness as tumor-selective PDT platforms. Different lipid conformations exhibit significant differences in photodynamic generation, phototriggered release, and cell phototoxicity.
