Metastatic Spiradenocarcinoma Managed With PD-1 Inhibition

Jeffrey J. Wargo et al.

Summary: Spiradenomas are rare skin tumors that usually appear in early adulthood and can degenerate into aggressive spiradenocarcinomas. Treatment recommendations are currently based on case reports and expert opinion, with no established standard of care for metastatic disease. This article presents the first reported case of a metastatic spiradenocarcinoma managed with pembrolizumab, showcasing the potential of immunotherapy in treating this condition.

Comparing Copy Number Variations in a Danish Case Cohort of Individuals With Psychiatric Disorders

Xabier Calle Sanchez et al.

Summary: This population-based case-cohort study examined the association between recurrent genomic copy number variants (CNVs) and mental disorders, finding that CNVs were broadly associated with an increased risk of autism spectrum disorder and ADHD, while the risk estimates for schizophrenia were lower. Sex may modulate the pathogenicity of many CNVs.

Risk of carcinogenesis in the biliary epithelium of children with congenital biliary dilatation through epigenetic and genetic regulation

Hiroki Mori et al.

Summary: The Ki-67 labeling index and expressions of KRAS, p53, HDAC, and AID in the gallbladder epithelium were significantly higher or tended to be higher in both children with CBD and adults with PBM compared to controls. This suggests that BTC may develop later in life through epigenetic and genetic regulation involving HDAC and AID expression.

Identification of variants in ACAN and PAPSS2 leading to spondyloepi(meta)physeal dysplasias in four Chinese families

Yixuan Cao et al.

Summary: This study identified novel and reported variants associated with spondyloepiphyseal dysplasias in four Chinese families, enriching the genetic background of skeletal disorders and expanding the mutation spectra of ACAN and PAPSS2. The pathogenicity of these variants was confirmed through conservation analysis, protein modeling, and bioinformatic predictions.

Gene networks and transcriptional regulators associated with liver cancer development and progression

Tatiana Meier et al.

Summary: The study aimed to improve understanding of HCC development and progression by combining different methodological approaches. Analysis revealed different gene regulatory networks underlying distinct biological features in HCC, with chromosomal instability being a common characteristic. New candidate genes for liver cancer were identified, and the necessity of different treatment approaches for low and high proliferation HCC forms was highlighted.

RIPK3 activation induces TRIM28 derepression in cancer cells and enhances the anti-tumor microenvironment

Han-Hee Park et al.

Summary: The study identifies TRIM28 as a co-repressor regulating transcriptional activity during necroptosis. Activated RIPK3 phosphorylates TRIM28, inhibiting its chromatin binding activity and contributing to increased cytokine expression and immunoregulatory processes. RIPK3 activation-dependent derepression of TRIM28 in cancer cells leads to enhanced immunostimulatory cytokine production and robust anti-tumor immunity.

Notch-Hes1 signaling activation in Caroli disease and polycystic liver disease

Kenta Takahashi et al.

Summary: The Notch signaling pathway plays a key role in the progression of biliary cystogenesis in Caroli disease and polycystic liver disease, possibly through the nuclear overexpression of Notch1 and Hes1 in biliary epithelial cells. Cell proliferative activity is significantly increased in both diseases.

Molecular Characteristics of Choledochal Cysts in Children: Transcriptome Sequencing

Yong Lv et al.

Summary: This study utilized RNA sequencing and bioinformatic analyses to identify differentially expressed genes and key modules associated with clinical subtypes of choledochal cyst (CC). It was found that cystic CC and fusiform CC exhibit distinct molecular mechanisms, with metabolism-related genes dysregulated in cystic CC and immune-related genes dysregulated in fusiform CC. A total of 20 candidate hub genes were identified, including ERBB2 and WNT11, which were verified to have differential expression in CC patients. These findings provide insight into the transcriptome characteristics of CC and have potential implications for personalized treatment.

Genomic gain of RRS1 promotes hepatocellular carcinoma through reducing the RPL11-MDM2-p53 signaling

Pengbo Cao et al.

Summary: The study revealed that the oncogene RRS1 on chromosome 8q promotes HCC development by regulating ribosome biogenesis activity and impacting the RPL11-MDM2-p53 pathway. Higher expression levels of RRS1 predict poor clinical outcomes in patients, especially in those with intact p53. This provides new potential targets for the treatment of this malignancy.
