Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Tumor Cell Glycolysis-At the Crossroad of Epithelial-Mesenchymal Transition and Autophagy

Fabrizio Marcucci et al.

Summary: There is ample evidence that glycolysis can induce epithelial-mesenchymal transition (EMT) and autophagy in tumor cells. Furthermore, upregulation of glycolysis can result in a mixed mesenchymal/autophagic phenotype in tumor cells.

PFKFB4 is overexpressed in clear-cell renal cell carcinoma promoting pentose phosphate pathway that mediates Sunitinib resistance

Chenchen Feng et al.

Summary: PFKFB4 was found to be significantly overexpressed in tumors compared to normal kidney, associated with advanced tumor grade, stage and poor prognosis. PFKFB4 overexpression impaired cell proliferation, migration, and wound healing. Targeting PFKFB4 showed promise in overcoming resistance to Sunitinib.

Melanoma Cell Resistance to Vemurafenib Modifies Inter-Cellular Communication Signals

Claudio Tabolacci et al.

Summary: The study focused on secretome profiling of melanoma cells sensitive or resistant to the BRAFi vemurafenib, finding that vemurafenib-resistant melanoma cells can influence dendritic cell maturation and produce higher levels of pro-inflammatory cytokines. Proteomic and cytokine/chemokine secretion analyses suggest a possible involvement of the proteasome pathway in BRAFi resistance, with BRAFi-resistant cells displaying a more aggressive phenotype and increased production of specific proteins. Plasma levels of CD147/basigin and MMP-2 were observed to increase in melanoma patients at the time of treatment failure, supporting the hypothesis that CD147/basigin might play a role in BRAFi resistance.

PFKFB4 promotes lung adenocarcinoma progression via phosphorylating and activating transcriptional coactivator SRC-2

Jiguang Meng et al.

Summary: PFKFB4 is overexpressed in lung adenocarcinoma and interacts with SRC-2. Phosphorylated SRC-2 by PFKFB4 promotes cancer cell proliferation, migration, and invasion. The PFKFB4-SRC-2-CARM1 axis plays a crucial role in lung adenocarcinoma progression.

Role of PFKFB3 and PFKFB4 in Cancer: Genetic Basis, Impact on Disease Development/Progression, and Potential as Therapeutic Targets

Krzysztof Kotowski et al.

Summary: Glycolysis is a crucial metabolic process in rapidly proliferating cells, such as cancer cells, with PFK-1 being a key rate-limiting enzyme regulated primarily by F-2,6-BP. PFK-2/FBPase-2 is a bifunctional enzyme responsible for F-2,6-BP synthesis and degradation, and alterations in PFKFB isozymes play important roles in various diseases, particularly in cancer.

Changing profiles of cancer burden worldwide and in China: a secondary analysis of the global cancer statistics 2020

Wei Cao et al.

Summary: This study summarized the recent changing profiles of cancer burden worldwide and in China, revealing that breast cancer became the most diagnosed cancer globally in 2020. China accounted for 24% of newly diagnosed cases and 30% of cancer-related deaths worldwide in 2020, with a high cancer mortality rate.

Bruceine A induces cell growth inhibition and apoptosis through PFKFB4/GSK3β signaling in pancreatic cancer

Pengfei Zhang et al.

Summary: Bruceine A exhibits strong anti-tumor activity against human pancreatic cancer cells in vitro and in vivo by targeting PFKFB4 and its downstream target GSK3 beta to inhibit glycolysis, thereby inducing cell growth inhibition and apoptosis. This suggests that bruceine A may serve as a potent agent for treating human pancreatic cancer.

E2F2 drives glioma progression via PI3K/AKT in a PFKFB4-dependent manner

Longzhou Zhang et al.

Summary: The study elucidated that E2F2 plays a crucial role in glioma malignancy progression by transcriptionally enhancing PFKFB4 expression and activating the PI3K/AKT pathway. Inhibition of E2F2 suppressed glioma growth and metastasis, indicating its potential as a therapeutic target.

PELP1/SRC-3-dependent regulation of metabolic PFKFB kinases drives therapy resistant ER+ breast cancer

Thu H. Truong et al.

Summary: Recurrence of metastatic breast cancer in women with luminal (ER+) breast cancer is a significant health burden, with contributing factors including the maintenance and expansion of breast cancer stem cells. The study demonstrates that cytoplasmic complexes composed of steroid receptor co-activators PELP1 and SRC-3 modulate breast cancer stem cell expansion by upregulating metabolic target genes PFKFB3 and PFKFB4. This suggests a potential new approach to overcoming endocrine and chemotherapy resistance in ER+ tumor cell populations.

Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

Daniel J. Klionsky et al.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
