Dithiocarbamates combined with copper for revitalizing meropenem efficacy against NDM-1-producing Carbapenem-resistant Enterobacteriaceae

Summary: The study constructed a compound library of dithiocarbamates and evaluated their synergistic antibacterial activities combined with copper. One compound was found to inhibit the growth and resistance development of NDM-1-producing Gram-negative pathogens, and a novel inhibition mechanism was uncovered.

Quercetin alleviates acute kidney injury by inhibiting ferroptosis

Summary: The study demonstrated that quercetin (QCT), a natural flavonoid, inhibited ferroptosis in renal proximal tubular epithelial cells by reducing lipid ROS levels and increasing glutathione (GSH) levels. QCT also ameliorated acute kidney injury induced by ischemia-reperfusion (I/R) or folic acid (FA). Furthermore, RNA-sequence analysis revealed ATF3 as a key factor in QCT's inhibition of ferroptosis. Additionally, QCT inhibited the recruitment of pro-inflammatory macrophages in AKI.

Interactions of the Diazabicyclooctane Serine β-Lactamase Inhibitor ETX1317 with Target Enzymes

Summary: ETX0282, an orally bioavailable prodrug of the diazabicyclooctane serine beta-lactamase inhibitor ETX1317, is currently being tested in combination with cefpodoxime proxetil for complicated urinary tract infections. ETX1317 contains a fluoroacetate moiety esterified with an isopropyl group in ETX0282 to provide high oral bioavailability. Studies on the interaction of ETX1317 with various beta-lactamases show that it has intermediate potency compared to other inhibitors like durlobactam and avibactam.

Unique Diacidic Fragments Inhibit the OXA-48 Carbapenemase and Enhance the Killing of Escherichia coli Producing OXA-48

Summary: This study screened various low-molecular-weight compounds against OXA-48 and identified functional scaffolds for inhibitor development with key interactions revealed. Some low-micromolar-potency inhibitors synergize with ampicillin to kill Escherichia coli expressing OXA-48, albeit at high concentrations of the respective inhibitors. Additionally, several compounds showed micromolar potency toward OXA-24 and OXA-58 class D carbapenemases prevalent in Acinetobacter baumannii.

Combining beta-lactam antibiotics with beta-lactamase inhibitors is the main strategy to overcome this issue. ID2 was identified as a potential inhibitor for OXA-48, demonstrating promising efficacy and lack of toxicity for further optimization programs.
