Protective Mechanism of Gandou Decoction in a Copper-Laden Hepatolenticular Degeneration Model: In Vitro Pharmacology and Cell Metabolomics

Summary: This study revealed the protective mechanisms of Gandou decoction (GDD) in a copper-laden hepatolenticular degeneration (HLD) model. GDD promoted copper excretion, improved ultrastructural changes in hepatocytes, decreased lipid peroxidation and reactive oxygen species levels, and increased antioxidant enzyme activity. Metabolomics analysis identified 47 biomarkers and 30 metabolic pathways related to GDD's pharmacological reactions in HLD model cells, with d-glutamate and d-glutamine metabolism showing the highest importance. Western blotting confirmed the differential expression of key proteins involved in these pathways. Overall, GDD protects hepatocytes by promoting copper excretion, regulating oxidative stress, and modulating protein expressions.
