Advances in clinical immunotherapy for gastric cancer

Kexin Li et al.

Summary: Gastric cancer is a highly malignant cancer with increasing incidence worldwide, ranking among the top five malignant tumors in terms of incidence and mortality. Immunotherapy, as a breakthrough in cancer treatment, has shown to be effective in gastric cancer treatment, especially when used in combination with other modalities.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

PD-L1, TMB, MSI, and Other Predictors of Response to Immune Checkpoint Inhibitors in Biliary Tract Cancer

Alessandro Rizzo et al.

Summary: ICIs have revolutionized the treatment landscape for several malignancies, but their efficacy in biliary tract cancer remains controversial. The lack of reliable predictive biomarkers presents a significant challenge in identifying responders to immunotherapy in this setting.

Tumor-Associated Macrophages and Inflammatory Microenvironment in Gastric Cancer: Novel Translational Implications

Karim Rihawi et al.

Summary: Gastric cancer is the fifth most commonly diagnosed cancer worldwide, with poor prognosis in advanced stage patients despite improvements in treatment. Research efforts have focused on identifying novel therapeutic targets, with the tumor microenvironment emerging as a potential target. Various agents and strategies targeting the TME are currently being evaluated in preclinical and clinical studies.

Gastric Cancer: Advances in Carcinogenesis Research and New Therapeutic Strategies

Lornella Seeneevassen et al.

Summary: Gastric cancer poses a significant global health issue due to its high incidence, poor prognosis, and cellular and molecular heterogeneity. Recent scientific advances have focused on understanding the different factors involved in gastric carcinogenesis, such as Helicobacter pylori, cancer stem cells, tumor microenvironment, and microbiota. Additionally, research has explored new potential biomarkers and emerging therapeutic strategies, including targeting cancer stem cells and immunotherapy, to combat cancer relapse, metastasis, and poor prognosis.

Exploration of Lipid Metabolism in Gastric Cancer: A Novel Prognostic Genes Expression Profile

Zhen Xiong et al.

Summary: This study found that higher expression levels of AKR1B1, PLD1, and UGT8 genes were associated with worse prognosis in GC patients, while AGPAT3 gene was associated with better prognosis. A gene expression profile consisting of AGPAT3, AKR1B1, PLD1, and UGT8 was developed, which stratified patients into three groups with significant overall survival differences. The profile was successfully validated in an independent cohort and performed well in an immunohistochemical cohort. Additionally, alterations in lipid metabolism were observed in GC, with upregulation of ether lipid metabolism, glycerophospholipid metabolism, and glycerolipid metabolism, as well as downregulation of fatty acid beta-oxidation and other lipid peroxidation processes.
