Hepatic fibrosis: 2022 Unmet needs and a blueprint for the future

Scott L. Friedman et al.

Summary: Steady progress has been made in understanding the pathogenesis and clinical consequences of hepatic fibrosis, but effective antifibrotic drugs are yet to be approved. This article highlights the successes and identifies gaps and unmet needs in both experimental and clinical aspects. Clarifying the mechanisms of fibrosis regression and developing better models will expedite drug development, while improving diagnostics and utilizing digital pathology methods can enhance prognostic information and better quantify therapeutic responses.

Transforming growth factor β latency: A mechanism of cytokine storage and signalling regulation in liver homeostasis and disease

Yujia Li et al.

Summary: Transforming growth factor-beta (TGF-beta) is an important factor in the liver that plays a role in various processes initiated by liver injury. It affects different types of liver cells and the response depends on the specific context. In the healthy liver, TGF-beta is stored in the extracellular matrix as latent TGF-beta (L-TGF-beta), and the protein ECM1 is critical for maintaining its latency. Activation of L-TGF-beta has detrimental effects on liver structure and function. This review article presents current knowledge on the formation, secretion, matrix deposition, and activation of L-TGF-beta, and highlights the potential therapeutic benefits of reducing L-TGF-beta activation in liver fibrosis and liver cancer.

Xiaoyaosan decoction alleviated rat liver fibrosis via the TGFβ/Smad and Akt/FoxO3 signaling pathways based on network pharmacology analysis

Yuan Zhou et al.

Summary: This study aimed to investigate the anti-fibrosis effect of Xiaoyaosan decoction (XYS) and explore the molecular mechanisms using network pharmacology and transcriptomic technologies. The results showed that XYS significantly improved liver function and alleviated fibrotic changes, possibly through the TGFβ1/Smad and Akt/FoxO signaling pathways.

Traditional Chinese Medicine Ganshuang Granules Attenuate CCl4-Induced Hepatic Fibrosis by Modulating Gut Microbiota

Jie Zhao et al.

Summary: This study demonstrated that ganshuang granules (GS) can alleviate liver injury and hepatic fibrosis induced by CCl4, by reducing oxidative stress and inflammation, and rebalancing gut microbiota. GS treatment enhanced tight junction-associated proteins in intestinal mucosa and improved the diversity of gut microbiota, showing promising potential in mitigating liver fibrosis.

Si-Wu-Tang ameliorates fibrotic liver injury via modulating intestinal microbiota and bile acid homeostasis

Xiaoyong Xue et al.

Summary: Si-Wu-Tang (SWT) showed significant therapeutic effects against carbon tetrachloride-induced liver fibrosis, improving collagen accumulation in the liver, intestinal barrier injury, and hepatic and intestinal inflammatory responses. By restructuring gut microbiota, influencing bile acid homeostasis, and activating the FXR pathways, SWT effectively protected against fibrotic liver injury induced by CCl4.

Targeting Nestin+ hepatic stellate cells ameliorates liver fibrosis by facilitating TβRI degradation

Huaxin Chen et al.

Summary: The study found that Nestin plays a significant role in liver fibrosis, with its expression being influenced by the TGFb-Smad2/3 signaling pathway and correlated with the degree of liver fibrosis. Knocking down Nestin reduced the activation of the TGFb pathway and fibrosis marker expression, thereby alleviating liver fibrosis.

TGFβ2 and TGFβ3 isoforms drive fibrotic disease pathogenesis

Tianhe Sun et al.

Summary: Research suggests that TGF beta 2 and TGF beta 3 may play a pathological role in fibrosis by showing increased expression in fibrotic tissues. Experimental evidence also confirms their independent involvement in mouse fibrosis models, with selective inhibition of TGF beta 2 and TGF beta 3 not leading to increased inflammation.

Ursolic acid reverses liver fibrosis by inhibiting NOX4/NLRP3 inflammasome pathways and bacterial dysbiosis

Yuan Nie et al.

Summary: The study found that UA can alleviate liver fibrosis by decreasing the expression of NOX4 and NLRP3, indicating their important role in liver fibrosis. Additionally, liver damage and fibrosis-related factors were significantly reduced in NLRP3(-/-), NOX4(-/-), and AP-treated mice after CCI4 injection, with a noticeable inhibition of NLRP3 expression in NOX4(-/-) and AP-treated mice.
