Differential expression of miR-148b, miR-129-2 and miR-296 in animal models of schizophrenia-Relevance to NMDA receptor hypofunction

Sowmya Gunasekaran et al.

Summary: Hypofunction of N-methyl-D-aspartate receptors (NMDAR) is a key component in the pathophysiology of schizophrenia. Alterations in the regulation of NMDARs by microRNAs (miRNAs) are possible since numerous miRNAs are differentially expressed in post mortem schizophrenia brain samples. Screening miRNAs altered in schizophrenia against NMDAR subunits Grin2A and Grin2B, it was found that miR-296, miR-148b, and miR-129-2 interacted with the 3' UTR sequences of both subunits. In vitro studies also showed that these miRNAs downregulated the expression of GluN2A and GluN2B proteins. Animal models of schizophrenia further supported these findings, with upregulation of miR-296-3p, miR-148b5p, and miR-137-3p, resulting in decreased GluN2A and GluN2B expression and altered expression of other schizophrenia-related genes.

Functional implications of CaMKII alternative splicing

Roman Sloutsky et al.

Summary: Ca2+/calmodulin-dependent protein kinase II (CaMKII) is a crucial regulator in the post-synapse during long-term potentiation, encoded by four genes in humans with alternative splicing leading to a diversity of proteins. Advances in sequencing technologies have led to the discovery of new CaMKII transcripts and suggest the adoption of a new naming scheme for CaMKII variants to better understand their biological implications.

NMDA receptor-independent LTP in mammalian nervous system

Karim A. Alkadhi

Summary: Long-term potentiation (LTP) is a form of activity-dependent synaptic plasticity that is closely associated with learning and memory. In the central nervous system, the activation of NMDAR is believed to be the main mechanism causing LTP, although other NMDAR-independent forms of LTP also exist.

Gating of hippocampal rhythms and memory by synaptic plasticity in inhibitory interneurons

Xingzhi He et al.

Summary: This study shows a positive correlation between hippocampal oscillatory strength and excitatory monosynaptic drive onto inhibitory neurons in freely behaving mice. By manipulating experience-dependent E -> I synaptic input/plasticity, the researchers were able to demonstrate its role in memory function. Deleting a specific protein affected long-term memory and brain wave rhythms.

The role of PKMζ in the maintenance of long-term memory: a review

Summary: This paper reviews the role of PKM zeta in the maintenance of long-term memory, highlighting its mechanism in preventing AMPA receptor endocytosis for synaptic potentiation. While inhibition of PKM zeta can reverse LTP and impair established long-term memories, a deficit in memory retrieval cannot be ruled out.

Calcium/Calmodulin-Dependent Protein Kinase II in Cerebrovascular Diseases

Xuejing Zhang et al.

Summary: Cerebrovascular disease, often leading to stroke, involves the multifunctional CaMKII protein. Past studies mainly focused on its role in neurons, but gaps remain in understanding how it functions in complex neurovascular processes. Emerging evidence suggests potential roles of CaMKII in the ischemic brain, sparking renewed research interest.

Conserved and divergent features of neuronal CaMKII holoenzyme structure, function, and high-order assembly

Olivia R. Buonarati et al.

Summary: By utilizing single-particle electron microscopy, a comparative analysis of neuronal CaMKII holoenzymes' alpha and beta isoforms was conducted, revealing different assembly states in the basal condition. The study also suggests that the beta isoform of CaMKII adopts a larger extended state, allowing for direct resolution of intra-holoenzyme kinase domain dimers.

Synaptic memory requires CaMKII

Wucheng Tao et al.

Summary: The study found that Ca2+-calmodulin-dependent kinase II (CaMKII) contributes to synaptic transmission and is essential for the maintenance of long-term potentiation (LTP). Acute inhibition of CaMKII erases LTP, while transient inhibition enhances subsequent LTP, strongly supporting the role of CaMKII as a molecular storage device.

The Calcium/Calmodulin-Dependent Kinases II and IV as Therapeutic Targets in Neurodegenerative and Neuropsychiatric Disorders

Kinga Salaciak et al.

Summary: CaMKII and CaMKIV are calcium/calmodulin-dependent kinases that play crucial roles in memory, plasticity, and various cellular functions. While they are primarily known for their involvement in memory processes, their effects on mood and neuropsychiatric diseases etiology are not well understood. This review aims to explore the structure and functions of these kinases and how they modulate behavior in animals to promote antidepressant-like, anxiolytic-like, and procognitive effects. The insights gained from this review can be beneficial in understanding the roles of these kinases in neurodegenerative and neuropsychiatric disorders and in future drug development.

L-Type Ca2+ Channels of NG2 Glia Determine Proliferation and NMDA Receptor-Dependent Plasticity

Na Zhao et al.

Summary: NG2 glia are essential proliferating cells in the CNS that can differentiate into oligodendrocytes and regulate homeostasis through secretion of growth factors and cytokines. L-type VGCCs in NG2 glia play a crucial role in cell proliferation and proper structural organization of nodes of Ranvier, while not affecting differentiation and myelin compaction. Additionally, L-type VGCCs contribute to the regulation of long-term neuronal plasticity.

GluN2B S1303 phosphorylation by CaMKII or DAPK1: No indication for involvement in ischemia or LTP

Jonathan E. Tullis et al.

Summary: The GluN2BDCaMKII mutation abolishes phosphorylation by CaMKII and DAPK1, potentially mediating neuroprotection through disruption of phosphorylation. However, S1303 phosphorylation was not increased by excitotoxic insults in hippocampal slices or global cerebral ischemia induced by cardiac arrest and cardiopulmonary resuscitation in vivo. In hippocampal cultures, S1303 phosphorylation was induced by chemical LTD but not LTP stimuli.

CaMKII activation persistently segregates postsynaptic proteins via liquid phase separation

Tomohisa Hosokawa et al.

Summary: The authors demonstrate that CaMKII undergoes liquid-liquid phase separation triggered by calcium signaling, which leads to the reorganization of postsynaptic structures and potentially enhances synaptic transmission efficacy during memory formation. This process of phase separation, induced by calcium signals, may serve as an activity-dependent mechanism to crosslink postsynaptic proteins, contributing to synaptic reorganization associated with synaptic plasticity.

CaMKII holoenzyme mechanisms that govern the LTP versus LTD decision

Sarah G. Cook et al.

Summary: The decision between long-term potentiation (LTP) and depression (LTD) in higher brain functions is determined by the complex cross-regulation of T286 and T305/306 autophosphorylation within the 12meric CaMKII holoenzyme. T305/306 phosphorylation selectively promotes LTD while T286 phosphorylation is required for both LTP and LTD. The differential T305/306 phosphorylation directs CaMKII movement to either excitatory or inhibitory synapses, coordinating plasticity at both synapse types.
