Transforming mutations in the development of pathogenic B cell clones and autoantibodies*

Summary: Autoimmune diseases are characterized by the presence of serum autoantibodies, which can be pathogenic and cause severe manifestations and organ injury. The development of these autoantibodies is driven by various genetic and environmental factors, but the factors behind the development of pathogenic autoantibodies are not well understood. Recent advancements in single-cell technology have allowed for the analysis of rare B cell clones producing pathogenic autoantibodies, revealing genetic events and mutations that disrupt antibody quality control mechanisms and decrease autoantibody solubility. Understanding the molecular characteristics and tolerance checkpoints involved in the formation of these pathogenic autoantibodies will improve prognostication, enabling early treatment to prevent organ damage and B cell malignancy.

Summary: Eligibility for trials in myasthenia gravis (MG) is mostly determined by a positive autoantibody serostatus, which limits potentially beneficial new treatments for seronegative MG (SNMG) patients. However, a subset of SNMG patients have detectable acetylcholine receptor (AChR) autoantibodies through clustered AChR cell based assay (CBA) testing. These findings support the clinical need for clustered AChR CBA testing in evaluating SNMG patients.

Convergent antibody responses to the SARS-CoV-2 spike protein in convalescent and vaccinated individuals

Summary: Unrelated individuals can produce genetically similar clones of antibodies known as public clonotypes, which have been identified in responses to different infectious diseases and in healthy individuals. By studying memory B cells from convalescent survivors of SARS-CoV-2 infection and plasmablasts from vaccinated individuals, researchers have identified 29 public clonotypes with potential neutralizing abilities against the virus. These findings provide insight into the molecular mechanisms behind the efficacy of SARS-CoV-2 vaccines and reveal the immune pressures influencing the selection of common viral escape mutants.
